摘 要:目的通过观察肠炎清对免疫复合溃疡型结肠炎模型(ICUC)大鼠结肠组织Toll样受体4(TLR 4)和核因子-κB(NF-κB)表达的影响,探讨肠炎清治疗IBD的作用机制。方法采用三硝基苯磺酸(TNBS)加免疫复合法造模。将大鼠随机分为空白对照组、模型对照组、肠炎清低、中、高剂量组(8.33、16.67、33.33m L/kg)、柳氮磺吡啶(SASP)组(0.5g/kg)。造模后第2 d,用药组分别给予相应药物灌胃7d,给药7天后麻醉处死大鼠,取新鲜结肠标本,采用免疫组化法检测TLR 4、NF-κBp65的表达,实时定量PCR法(qRT-PCR)检测TLR 4mRNA的表达。结果免疫组化结果:NF-κBp65和TLR4在各组各只动物均可见阳性表达,其中肠炎清高剂量组及SASP组可显著下调NF-κBp65的表达,有统计学差异(P<0.05)。TLR4在各给药组与模型组比较未见显著差异(P>0.05)。PCR结果:与模型对照组相比,各给药组大鼠结肠组织TLR4表达均为阳性,但未见统计学差异(P>0.05)。结论下调TLR4、NF-κBp65的表达,是肠炎清治疗IBD的作用机制之一。Objective : To observe the effect of Changyanqing on the colon tissue Toll - like receptor 4 ( TLR 4) and nuclear factorκB ( NF - κB) expression of immune complexes ulcerative colitis (ICUC) rats and explore its mechanism to treat IBD. Methods : Trinitro- benzene sulfonic acid (TNBS) plus immune complex was used to make models. Rats were randomly divided into a control group, a model control group, Changyanqing groups with low, medium and high dose (833, 1667, 3333mL/kg) and sulfasalazine (SASP) group (0.5g/ kg). In the following the day after modeling, the treatment groups were administered with corresponding drugs for 7 days, and the rats were sacrificed 7 days later to take their fresh colonic samples. Immunohistochemistry was used to detect TLR 4 and NF - κBp65 expression, and RT - PCR method to detect TLR 4mRNA expression. Results : Immunohistochemical results : NF - κBp65 and TLR4 in each group can be seen positive expression, among which the expression of NF - κBp65 of Changyanqing group with high dose and SASP group could be significantly down - regulated, and there are significant differences ( P 〈 005). TLR4 in each treatment group and the model group showed no significant difference (P 〉 005). PCR Results, compared with the model group, TLR4 expressions of the colon tissue of each adminis- tration group were positive, but there was no significant difference (P 〉 005). Conclusion: Changyanqing can down - regulate TLR4, NF - κBp65 expression, and its mechanism may be related to the regulation of TLR4 / NF - κB signal pathway.
