壮骨止痛方对去卵巢骨质疏松大鼠下丘脑瘦素及其受体的影响

摘　　要：目的观察壮骨止痛方对去卵巢骨质疏松大鼠下丘脑瘦素及其受体的影响,探讨其抗骨质疏松机制。方法 72只200 g左右的SD大鼠按体质量随机抽出假手术组12只,造模组60只,造模组采用双侧去卵巢法造模,假手术组只切除卵巢周围相应质量的脂肪。造模成功后,造模组大鼠按体质量随机分为模型组、壮骨止痛方高剂量组(13.2 g/kg)、中剂量组(6.6 g/kg)、低剂量组(3.3 g/kg)和戊酸雌二醇组,每组12只。术后第5天开始药物干预,模型组和假手术组给予相应体积的纯净水,持续13周。给药结束后,检测大鼠骨密度和下丘脑Leptin含量及OB-Rb蛋白表达。结果模型组大鼠下丘脑Leptin及OB-Rb含量显著降低,骨密度显著减少,与假手术组比较差异有统计学意义。壮骨止痛方各剂量组大鼠OB-Rb蛋白表达显著增加,骨密度显著增加,与模型组比较差异有统计学意义。结论促进OB-Rb蛋白表达,可能是壮骨止痛方抗骨质疏松的作用机制之一。Objective To explore the effect of Zhuanggu Zhitong prescription on the Leptin and OB-Rb in the hypothalamus of ovariectomized rats with osteoporosis, and to investigate its mechanism in anti-osteoporosis. Method 72 rats were randomly divided into sham group （12） and model group （60）. The model group rats were ovarieetomized, and the sham group rats were excised equal weight fat. After the modeling, the rats were randomly divided into model group, Zhuanggu Zhitong prescription high-dose group, Zhuanggu Zhitong prescription middle-dose group, Zhuanggu Zhitong prescription low-dose group and estradiol valerate group, with 12 rats in every group. After 5 days of the operation, rats started to receive drug therapy for 13 weeks. The model group and the sham group rats were given equal volume water. After the test, the content of Leptin and OB-Rb in the hypothalamus was detected. Results The content of Leptin and OB-Rb in the hypothalamus of ovariectomized rats with osteoporosis was significantly declined （ P 〈 0. 05）. After drug therapy, the content of OB-Rb in the hypothalamus and BMD of ovariectomized rats with osteoporosis were significantly increased （ P 〈 0.05 ）. Conclusion Promoting the expression of OB-Rb may be one mechanisms of Zhuanggu Zhitong prescription in anti-osteoporosis.
