摘  要:目的:观察青光安颗粒剂4种有效组分对兔眼滤过术后滤过道瘢痕组织TGF-β1和Smad3表达的影响,探讨青光安有效组分抗青光眼术后滤过道瘢痕化的效果及作用机制。方法:将青光安颗粒剂有效组分作用于滤过性手术后兔眼(有效组分1-4组),通过与正常组、模型组、丝裂霉素组(MMC组)的比较,观察青光安颗粒剂4种有效组分对青光眼术后滤过道瘢痕组织TGF-β1和Smad3的影响。结果:有效组分2组与MMC组TGF-β1和Smad3及m RNA表达比较差异无统计学意义,其余3组与MMC组TGF-β1和Smad3及m RNA表达比较,差异有显著性(P<0.05),其抑制效果不如MMC组。结论:青光安有效组分2和MMC都可通过抑制TGF-β1和Smad3 m RNA及其蛋白表达减少瘢痕组织增生,具有明显的抗青光眼术后滤过道瘢痕化的作用。Objective: To observe the effects of four active ingredients of Qingguang'an Granules on the expression of TGF-β1 and Smad3 in scarring tissues of filtering tract after filtering surgery in rabbits, and explore its mechanism underlying antiscarring of active ingredients. Methods: All rats were randomly divided into the control group(A group), model group(B group), Mitomycine C group(C group), and four active ingredients of Qingguang'an Granules groups abbreviated including D group, E group, F group and G group. The expression of TGF-β1 and Smad3 in scarring tissues of filtering tract after filtering surgery in rabbits were observed to evaluate the effects of four active ingredients of Qingguang'an Granules. Results: Expressions of TGF-β1 and Smad3 of No.2 active ingredient group were not obviously different from those of group C. At the same time expressions of TGF-β1 and Smad3 of the other three active ingredient groups had significant difference, comparing with those of MMC group(P0.05), and the inhibitory effect of three active ingredient groups were inferior to that of MMC group. Conclusion: Both No.2 active ingredient and MMC could inhibit expressions of TGF-β1 and Smad3 to decrease formation of scar tissues, which had obvious antiscarring of filtering tract after surgery.
