基于冠心病血瘀证心肌细胞能量代谢网络模型的影子价格分析

摘  要:目的:探讨冠心病(CHD)急、慢性血瘀证心肌细胞能量代谢的变化特点。方法:1运用Flux Explorer软件平台构建起的CHD血瘀证心肌细胞能量代谢网络模型;2应用GC-MS技术和血气自动分析仪从健康对照组、CHD慢性血瘀证、CHD急性血瘀证大鼠模型中取冠状动脉血检测葡萄糖、脂肪酸及血氧含量;3检测结果代入网络模型,通过影子价格分析CHD不同血瘀证状态下参数变化的特点。结果:在健康对照、CHD慢性血瘀证和CHD急性血瘀证的状态下,葡萄糖、脂肪酸、血氧的影子价格依次是-4.3478,0.0000,-5.6087;若分别增加1单位,则代谢后ATP的产量依次增加4.3478、0.0000、5.6087单位。3个模型中对于葡萄糖、脂肪酸和血氧3种能量物质的灵敏度有别,其中对于葡萄糖和血氧的灵敏度,呈现健康对照>CHD慢性血瘀证>CHD急性血瘀证的趋势(P<0.01);而对于脂肪酸,则呈健康对照<CHD慢性血瘀证<CHD急性血瘀证的趋势(P<0.01)。结论:CHD急性血瘀证和慢性血瘀证心肌细胞对于不同能量物质的利用程度有别,而提供充足的氧气是充分利用能量物质的关键。Objective: To study the characteristics of myocardial cell energy metabolism changes of acute and chronic blood stasis syndrome of coronary heart disease (CHD). Methods: ①Using FluxExplorer software platform to build up the CHD, myocardial cell energy metabolic network model of blood stasis syndrome. ②The coronary arterial blood glucose, fatty acids and blood oxygen content in model rats from groups of healthy control, CHD with chronic blood stasis syndrome, CHD with acute blood stasis syndrome were detected by GC-MS technology and automatic blood gas analyzer. ③The results were brought into network model, and characteristics of parameters in different blood stasis syndrome of CHD were analyzed by shadow price. Results: In the condition of healthy subjects, CHD with chronic blood stasis syndrome, and CHD with acute blood stasis syndrome, the shadow price of glucose, fatty acid, oxygen were -4.3478, 0.0000, -5.6087. If increased 1 unit respectively, the metabolism of the ATP after yield increased by 4.3478, 0.0000, 5.6078 units. The sensitivity of three kinds of energy substances (glucose, fatty acid and oxygen) in three models were vary, which for glucose and oxygen sensitivity, the trend was healthy control〉CHD with chronic blood stasis syndrome〉CHD with acute blood stasis syndrome (P〈0.01); But for the sensitivity of fatty acid, the trend was healthy control〈CHD with chronic blood stasis syndrome〈CHD with acute blood stasis syndrome (P〈0.01). Conclusion: The utilization of different energy substances of myocardial cells in CHD with chronic blood stasis syndrome and CHD with acute blood stasis syndrome are different, and to provide sufficient oxygen is the key to make full use of energy substances.
