摘 要:目的:观察细胞外信号调节激酶(ERK)、Wnt/β-连环蛋白(β-catenin)信号通路相关蛋白在右归饮含药血清影响成骨细胞过程中的作用,明确右归饮含药血清对ERK、Wnt通路和ERK/Wnt交叉串扰的作用。方法:体外培养成骨细胞,分组,加药干预。培养3d后,应用ELISA检测Wnt/β-catenin及ERK通路相关蛋白β-catenin、Runx2、ERK蛋白表达情况。结果:右归饮含药血清能显著上调3种蛋白表达(P<0.05,P<0.01)。联合阻断ERK、Wnt通路,3种蛋白表达水平较含药血清组有明显降低(P<0.05,P<0.01);与单独阻断其中一条通路比较,有明显下降(P<0.01)。单独阻断某一条通路,仅Runx2较右归饮含药血清组有明显降低(P<0.05,P<0.01)。结论:右归饮含药血清能显著增加成骨细胞活性,该作用与ERK、Wnt信号通路密切相关,且两条通路之间存在一定的交叉串扰作用。Objective: To investigate the influence of Wnt/β-catenin and ERK signal pathway in the effect of Youguiyin containing serum on osteoblast, and to elucidate the effects of Youguiyin containing serum on ERK, Wnt/β-catenin signal pathway and the crosstalk between the two pathways. Methods: The osteoblast(OB) from cranial bones of rat neonates were isolated and cultured for 3 days in vitro, which were divided into 5 groups. Levels of relevant proteins in Wnt/β-catenin and ERK signal pathway including β-catenin, ERK and Runx2 in each group were detected with ELISA assay. Results: Youguiyin containing serum could significantly upregulate the expression of β-catenin, ERK, Runx2 protein(P〈0.05, P〈0.01). When ERK and Wnt/β-catenin signal pathway were blocked simultaneously, the level of β-catenin, ERK, Runx2 protein reduced significantly compared to the Youguiyin containing serum group(P〈0.05, P〈0.01). The level of β-catenin, ERK, Runx2 protein reduced significantly compared to thesituation when either of the pathways was blocked. When block ERK signal pathway alone or Wnt/β-catenin signal pathway alone, only the level of Runx2 reduced significantly compared to the Youguiyin containing serum group(P〈0.05, P〈0.01). Conclusion: Youguiyin containing serum could apparently increase activity of osteoblast. This action was closely correlated with ERK signal pathway and Wnt/β-catenin signal pathway. Meanwhile there must be some crosstalk effects between the two pathways.
