摘 要:目的:探讨鳖甲煎丸不同提取物对转化生长因子-β1(TGF-β1)诱导的HSC-T6细胞增殖及相关细胞因子的影响作用,并探究水提物与醇提物抗肝纤维化作用的异同。方法:建立TGF-β1诱导HSC-T6细胞增殖模型,采用MTT法观察水提物与醇提物对HSC-T6细胞增殖的量效和时效关系来探讨鳖甲煎丸提取物对TGF-β1诱导HSC-T6细胞增殖的影响;并采用RT-PCR、ELISA法测定不同提取物对HSC-T6细胞相关细胞因子的影响。结果:鳖甲煎丸水提物在0.125-2.000mg/m L浓度范围,抑制效果随剂量增加而降低,呈线性负相关关系,醇提物未见明显的剂量依赖关系。水提物抑制作用随时间延长表现为先增强后减弱,醇提物则表现出一定的时间依赖性,随时间延长而增强。与空白对照组比较,模型组细胞相关细胞因子α-SMA、Col-Ⅲ、CTGF和PDGF m RNA含量增高(P<0.05)。与模型组比较,鳖甲煎丸水提物使HSC-T6细胞COL-Ⅲ、COL-Ⅳ、PDGF m RNA含量和蛋白表达降低。醇提物使HSC-T6细胞COL-Ⅲ、COL-Ⅳm RNA含量和蛋白表达降低,而PDGF m RNA含量升高促进其蛋白表达。结论:鳖甲煎丸水提物与醇提物对TGF-β1诱导的HSC-T6细胞增殖均有一定的抑制作用,但其抑制作用表现有一定差异;同时均可抑制胶原COL-Ⅲ、COL-Ⅳ的合成;对PDGF表达调控亦有所不同。Objective: To investigate the effects of different extracts from Biejiajian Pill on the proliferation and relevant cytokines of TGF-β1 induced HSC-T6 cells, and to explore the difference between the effects of aqueous and ethanol extracts on the anti-hepatic fibrosis. Methods: TGF-β1 was used to induce HSC-T6 cell to establish the proliferation model. MTT assay was used to observe the dose-effect and time-effect of extracts on the proliferation of HSC-T6 cells induced by TGF-β1. The relevant cytokines were detected by RT-PCR and ELISA assay. Results: Aqueous extract showed a significant negative linear correlation between dose and inhibition effect in the 0.125-2.000mg/m L concentration range, while ethanol extract had no obvious doseeffect relationship. With the time prolongs, the inhibition effect of aqueous extractwas strengthened first and then weakened. Ethanol extract had a time-effect relationship, which was strengthenedwith the time. Compared with the blank control group, model group increased the expression of α-SMA, COL-Ⅲ, CTGF, PDGF m RNA(P〈0.05). Compared with the model group, aqueous extract reduced the expression of COL-Ⅲ, COL-Ⅵ, PDGF. Ethanol extract reduced the expression of COL-Ⅲ, COL-Ⅵ, but increased PDGF. Conclusion: Both aqueous extract and ethanol extract of Biejiajian Pill have an inhibitory effect on the proliferation of TGF-β1 induced HSC-T6 cell, and both the aqueous extract and ethanol extract of Biejiajian Pill can inhibit expressions of COL-Ⅲ and COL-Ⅵ, but have different performances to PDGF.
