摘  要:目的探讨渐尖毛蕨总黄酮对前列腺增生大鼠的保护作用。方法大鼠随机分为假手术组8只和造模组,造模组手术切除双侧睾丸,手术成功操作后按照随机数字表法分为4组:模型对照组、非那雄胺组、总黄酮大剂量组和总黄酮小剂量组,每组8只。造模大鼠皮下注射10 mg·kg^(-1)·d^(-1)丙酸睾酮(溶于橄榄油),假手术组皮下注射等体积橄榄油。注射1 h后,非那雄胺组灌胃非那雄胺5 mg·kg^(-1)·d^(-1),总黄酮大、小剂量组分别给予总黄酮200和100 mg·kg^(-1)·d^(-1),模型对照组和假手术组灌胃等体积0.9%氯化钠溶液。观察各组大鼠前列腺组织形态学变化、抗氧化酶活力、炎性细胞因子水平、前列腺生长因子和microRNA(miR)27a/b表达程度。结果与假手术组比较,模型对照组大鼠前列腺指数(PI)和血清前列腺酸性磷酸酶(PACP)活力明显上升(均P<0.01),超氧化物歧化酶(SOD)和谷胱甘肽过氧化酶(GPx)活力显著下降,丙二醛(MDA)水平大幅上升(均P<0.01),白细胞介素(IL)-1β、IL-6、肿瘤坏死因子-α(TNF-α)和环氧合酶-2(COX-2)的水平均明显升高(均P<0.01),前列腺miR27a和miR27b表达明显上升(均P<0.01);与模型对照组比较,总黄酮大、小剂量组PI和PACP下降(P<0.01),SOD和GPx的酶活力明显上升(P<0.01),MDA水平降低(P<0.05),前列腺IL-1β、IL-6、TNF-α和COX-2升高的趋势显著抑制,显著抑制miR27a表达上升的趋势(P<0.01)。大剂量总黄酮能明显抑制miR27b的表达(P<0.05)。结论渐尖毛蕨总黄酮具备潜在的抗前列腺增生活性。Objective To assess the potential of total flavonoids from Cyclosorusacuminatus (TFC) to protect against benign prostatic hyperplasia in rats. Methods Rats were randomly divided into sham-operation group(n=8) and model control group.Model control group was castrated and then divided into four groups according to the random number table: the model control group,the finasteride group,the high and low doses TFC groups (n=8 each group).The rats from model groups were subcutaneously injected with 10 mg·kg^-1·d^-1 of testosterone (dissolved in olive oil).The sham-operation group was subcutaneously injected with the same volume of olive oil.One hour after the injection,the finasteride group was intragastrically administrated with 5 mg·kg^-1·d^-1 of finasteride.The high and low dose TFC groups were given 200, 100 mg·kg^-1·d^-1 of TFC.The sham-operation group and the model control group were given the same volume of 0.9% sodium chloride solution.The prostatic changes of morphology,activities of antioxidant enzymes,as well as the levels of inflammatory cytokines,growth factors and microRNA (miR) 27a/b were observed. Results Compared with the sham-operation group,the level of PI and the activity of serum PACP were significantly enhanced (P〈0.01),the activities of SOD and GPx were significantly decreased (P〈0.01),the prostatic levels of MDA,IL-1β,IL-6,TNF-α,COX-2,miR27a,and miR27b were significantly increased (P〈0.01) in the rats of model control group.Compared to the model control group,TFC significantly inhibited the increase of PI and PACP (P〈0.01),normalized the activities of SOD and GPx (P〈0.01),decreased the level of MDA (P〈0.05),suppressed the enhancement in the levels of IL-1β,IL-6,TNF-α and COX-2,and significantly inhibited the over expression of miR27a (P〈0.01).TFC (200 mg·kg^-1·d^-1)significantly inhibited the expression of miR27b (P〈0.05). Conclusion TFC is able to exert protective effect in benign prostatic hyperplasia rats.
