摘  要:目的:比较不同剂量亚氨基二丙腈(IDPN)对诱导的多发性抽动症(TS)大鼠模型的影响。方法:80只雄性SD大鼠随机分为空白对照组、低剂量组、中剂量组、高剂量组并分别予生理盐水、IDPN 150 mg/kg、IDPN300 mg/kg、IDPN 350 mg/kg腹腔注射,每日1次,连续7日,比较各组大鼠一般情况、刻板行为、运动行为、病理学形态及多巴胺(DA)含量变化。结果:较空白对照组,低剂量组一般情况好,未出现死亡,刻板行为、运动行为增多,病理学改变不明显,DA含量变化不明显;中剂量组一般情况好,死亡率低,刻板行为、运动行为明显增多,病理学形态改变不明显,DA含量变化不明显;高剂量组一般情况较差,死亡率高,刻板行为、运动行为显著增多,病理学形态改变明显,DA含量未见明显变化。结论:本实验成功建立了TS大鼠模型,三种剂量均可完成造模,中剂量组(300 mg/kg)可缩短造模时间,成功率高,死亡率低,能为TS的实验研究提供较好的可复制动物模型。Objective:To compare different doses of IDPN on induced tourette syndrome (TS) model in rats. Methods: Eighty male SD rats were randomly divided into blank control group, low dose group, middle dose group and high dose group. And they were given normal saline,IDPN 150 mg/kg, IDPN 300 mg/kg and IDPN 350 mg,/kg by intraperitoneal injection separately. Comparisons of general condition, stereotyped behavior, exercise behavior, pathology and the level of dopamine (DA) in each group were carried out. Result: Compared with blank control group, the general condition in the low dose group was well. The stereotyped behavior and exercise behavior increased and the pathology change was not obvious and the content of DA not obvious. The middle dose group's general condition was well. The stereotyped behavior and exercise behavior increased obviously and the pathology change was not obvious and DA content change not obvious with low mortality rate. The high dose group's general situation was bad. The stereotyped behavior and sports behavior increased significantly. The pathology changed significantly and DA content change was not obvious. The mortality was high in high dose group. Conclusion: Three different doses all can establish the TS rats models successfully. And the model of middle dose group (300 mg/kg) need short time,with high successful rate and low death rate, providing an optimal animal model for experimental study of TS.
