摘 要:目的:探讨中成药醒脑静注射液对老年麻醉后认知功能障碍大鼠学习记忆功能及海马区PS-1、TTBK-1表达的影响,研究其改善老年认知障碍大鼠学习记忆功能的可能机制。方法:将40只老年大鼠随机分为空白对照组10只,余30只制备老年认知障碍大鼠模型复制组,随机分为模型对照组10只、醒脑静低剂量组10只、醒脑静高剂量组10只,共4组。免疫组化法观察各组海马组织PS-1、TTBK-1的表达。结果:与空白对照组比较,老年认知障碍大鼠模型复制组逃避潜伏期显著延长、穿越平台次数显著减少(P<0.01),治疗给药6 d后,与空白对照组比较,模型对照组、醒脑静低剂量组、醒脑静高剂量组海马组织PS-1、TTBK-1表达显著增多(P<0.01);与模型组比较,醒脑静低剂量组、醒脑静高剂量组海马组织PS-1、TTBK-1表达显著减少(P<0.05,P<0.01)。结论:醒脑静注射液可降低老年认知功能障碍大鼠海马区PS-1、TTBK-1的表达,从而保护海马组织形态的完整性,对老年认知功能障碍大鼠起到较好的治疗作用。Objective:To explore the effect of Chinese medicine Xingnaojing Injection on postoperative cognitive impairment elderly rats' learning and memory function and the expressions of PS - 1 and TTBK - 1 in hippocampus and study the possible mechanisms of improving geriatric cognitive impairment rats' learning and memory function. Method: A total of 40 aged rats were randomly divided into blank control group( n = 10) and the rest 30 were established the elderly cognitive impairment models which were randomly divided into model control group( n=10) ,Xingdaojing Injection low dose group( n = 10) and Xingnaojing Injection high dose group (n = 10). Use Morris water maze to observe and evaluate the improvement of spatial learning and memory. Use Immunohistochemical method to observe expressions of PS - 1 and TFBK - 1 in each rats' hippocampus. Result.: Compared with the blank control group, elderly cognitive impairment models' escape incubation period was significantly prolonged and the platform - crossing times significantly reduced(P 〈 0. 01 ). After treatment for 6 days, compared with the blank control group, model control, Xingdaojing Injection low dose and Xingnaojing Injection high dose groups' expressions of PS - 1 and TTBK - 1 in hippocampus were increased significantly (P 〈 0.01 ). Compared with model group, Xingdaojing Injection low dose and Xingnaojing Injection high dose groups' expressions of PS - 1 and TTBK - 1 in hippocampus were significantly reduced(P 〈0.05 ,P 〈0. 01 ). Conclusion:Xingdaojing Injection can reduce the expressions of PS - 1 and TTBK - 1 of age - related cognitive dysfunction rats' hippocampus, and thus ensure the integrity of the hippocampus morphology and improve cognitive impairment in elderly rats' spatial learning and memory ability.
