摘  要:目的:探讨血府逐瘀汤治疗慢传输型便秘的机制。方法:建立大鼠慢传输型便秘模型,通过激光多普勒血流仪扫描活体大鼠结肠,比较大鼠结肠血流灌注量和血流速度;硝酸还原酶法测大鼠血清一氧化氮(NO),酶联免疫吸附试验法(ELISA)检测大鼠血清内皮素(ET)的水平。结果:模型组大鼠结肠微循环血流灌注量较其它组均明显升高,差异有统计学意义(P<0.05);给药后,低剂量组与中剂量组大鼠结肠黏膜微循环灌注量较模型组降低,但两者无差异(P>0.05),高剂量组与阳性组较中剂量组明显降低,但两者间无统计学意义(P>0.05)。模型组血清一氧化氮(NO)、血清内皮素(ET)含量均较其他组显著增高,差异有统计学意义(P<0.05);给药后,低剂量组与中剂量组均较模型组明显减低,但两组间无差异(P>0.05);高剂量组、阳性组血清ET/NO较中剂量组明显降低,但高剂量组与阳性组无明显差异,(P>0.05)。结论:慢传输型便秘的病因病机与气滞血瘀相关;血府逐瘀汤改善结肠黏膜微循环;血府逐瘀汤参与慢传输型便秘中的ET/NO机制,对慢传输型便秘有效,且与药量成正相关;血府逐瘀汤高剂量组与枸橼酸莫沙比利效果相似。Purpose:To study the therapeutic mechanism for the treatment of slow transit constipation by xuefu zhuyu decoction. Method: Build model in rats by scanning rats colon with Laser Doppler Flowmete, compared blood flow velocity and blood perfusion. Results: Blood flow velocity and blood perfusion were significantly higher than other groups, the difference was statistically significant (P 〈0. 05 ) ; After the treatment, the low dose group and the middle dose group reduced, but no difference(P 〉0.05) ,both high dose group and positive group are significantly lower, but no statistical significance (P 〉 0.05 ). Model group serum nitric oxide (NO), endothelin (ET) serum levels were significantly higher than other group, the difference was statistically significant (P 〈 0.05) ; After the treatment, the low dose group and the middle dose group reduced, but no difference ( P 〉 0. 05 ), both high dose group and positive group are significantly lower, but no statistical significance ( P 〉0.05 ). Conclusion: The etiology and pathogenesis of slow transit constipation related with qi and blood stasis; Xuefu zhuyu decoction improve colon mucosal microcirculation, to participate in ET/NO mechanism, while dosen is negatively correlated. The high dose group of Xuefu zhuyu decoction and Citrate moser Billy are similar effect.
