摘 要:目的研究蟾毒灵对人肠癌HCT116瘤体生长产生的抑制作用。方法将25只人肠癌移植瘤模型裸鼠以随机区组法分为空白组(生理盐水)、对照组(5-Fu,5 mg·kg-1·d^(-1))和3个剂量(蟾毒灵,0.5,1.0,1.5 mg·kg-1·d^(-1))实验组,均为每只0.2 m L,每周3次,共3周。以CCK-8法测定细胞增殖作用并记录瘤体生长情况;用免疫组化法测量Ki67蛋白的表达。结果蟾毒灵对HCT116细胞的增殖有明显的抑制作用,呈剂量和时间依赖性,其半数抑制浓度(IC50)为80 nmol·L^(-1),差异有统计学意义(P<0.01)。各组瘤体生长情况:与空白组(170±104.74)mg相比,低中高3个剂量实验组瘤重分别为(118.75±54.62),(107.5±41.31),(103.75±70.29)mg,差异均有统计学意义(均P<0.05);与对照组的(112.5±65.85)mg比较,差异也有统计学意义(均P<0.05)。免疫组化结果表明,蟾毒灵可以下调Ki67蛋白的表达水平。结论蟾毒灵可抑制人肠癌HCT116瘤体生长,可能与降低Ki67的表达有关。Objective To study the inhibition effect of Bufalin on HCT116 colon cancer cell proliferation and tumor growth.Methods Twenty-five subcutaneous xenograft model BALB/c mice were randomized into the blank group(0.9% normal saline),control group(5-Fu,5 mg·kg-1·d^(-1)) and low,medium,high doses test groups(Bufalin,0.5,1.0,1.5 mg·kg-1·d^(-1)).They were injected 3times every week,continuous administration of 3 weeks.Cell proliferation were determined by CCK-8.The tumor inhibition rate in mice were calculated.The expression of Ki67 was detected through immunohistochemistry.Results Bufalin had obvious inhibition on proliferation of HCT116 cell and showed a dose and time dependence,and IC50 was 80nmol· L^(-1)(P〈0.01).Tumor growth in each group was observed as follows:compared to blank group with(170 ± 104.74) mg,weight of tumor in low,medium,high doses test groups with(118.75 ± 54.62),(107.5 ± 41.31),(103.75 ± 70.29) mg,and the difference was statistically significant(all P〈0.05).And compared to control group with(112.5 ± 65.85) mg,the difference was also statistically significant(P〈0.05).That indicating that test group and control group had inhibits tumor growth more than blank group.Immunohistochemistry results revealed that high dosage group of Bufalin and control group can reducethe expression of Ki67.Conclusion Bufalin can inhibit proliferation and tumor growth of the HCT116 colon cancer cell,perhaps by the way to download the expression of Ki67.
