摘 要:目的探讨脂联素(APN)及其基因位点+45 T/G和-11377 C/G的多态性与广西地区壮族代谢综合征(MS)患者的相关性。方法 ELISA法检测广西地区壮族和汉族MS患者、壮族和汉族健康人群(各100例)血清APN水平,PCR-RFLP技术检测APN基因-11377 C/G和+45 T/G位点多态性。结果与同民族健康组比较,两民族MS组血清APN水平均降低(P均〈0.05)。4组受试者APN+45等位基因分布频率差异有统计学意义(χ2=26.560,P=0.000),且与同民族健康对照组比较TT和TG基因型分布频率降低,GG基因型升高(P均〈0.05)。4组受试者APN-11377位点等位基因分布频率差异无统计学意义(χ2=2.051,P=0.562)。此外,不同民族MS患者APN+45 T/G基因型间血清APN水平差异有统计学意义,且与同民族TT基因型比较,TG和GG型均降低(P均〈0.05)。结论 APN+45位点的多态性及血清APN水平改变可能与广西地区壮族和汉族MS发生密切相关。Objective To investigate the correlations between adiponectin( APN),APN + 45 and APN-11377 genotyping and metabolic syndrome( MS) in ethnic Zhuang patients. Methods The levels of serum APN in ethnic Zhuang and Han patients with MS as well as the healthy individuals( each of 100 cases) were measured by ELISA,and APN + 45 and APN-11377 genotyping were detected by using PCR-RFLP. Results Compared with the healthy control group,the APN levels of both MS groups were significantly decreased( P 〈 0. 05). The difference of allele frequency of APN + 45 gene polymorphisms in four groups was statistically significant( χ~2=26. 560,P = 0. 000). Compared with the normal control groups,TT and TG genotype frequency in two MS groups were significantly lower,GG genotype frequency were significantly higher( P 〈 0. 05). Among the 4 groups,the difference of APN-11377 allele frequency was not statistically significant( χ~2= 2. 051,P = 0. 562). The serum APN level in the MS patients with APN + 45 TT genotype was significantly higher than that of the patients with TG and GG genotype,and the difference was statistically significant( all P 〈 0. 05).Conclusion The polymorphism of the APN + 45 locus resulted in the change of serum APN levels may associate with the occurrence of MS in the population of Zhuang and Han nationality.
