摘  要:【目的】探讨小花棘豆中毒对家兔丘脑-垂体-性腺轴α-甘露糖苷酶(AMA)的影响,揭示小花棘豆的毒性作用机理。【方法】将24只家兔随机分为对照组和试验Ⅰ、Ⅱ、Ⅲ组,对照组饲喂青干草,试验Ⅰ、Ⅱ、Ⅲ组分别饲喂添加15%,30%和45%小花棘豆全草的混合饲料(其苦马豆素含量分别为30,60和90mg/kg),分别于攻毒后第14,35和70天时每组随机采集2只家兔的丘脑、垂体和性腺,检测AMA在家兔丘脑-垂体-性腺轴中的分布、活性及其基因mRNA的表达量。【结果】AMA在家兔丘脑、垂体和性腺中均有分布,小花棘豆中毒可导致家兔丘脑-垂体-性腺轴AMA活性和分布明显下降。试验组及对照组家兔丘脑-垂体-性腺轴高尔基体α-甘露糖苷酶Ⅱ(AMA1)和溶酶体α-甘露糖苷酶(AMA2)均有表达,但各试验组表达转录水平总体低于对照组,攻毒第70天时试验Ⅰ组家兔丘脑-垂体-性腺轴的AMA1和AMA2,试验Ⅱ组家兔丘脑-垂体-性腺轴及试验Ⅲ组家兔丘脑和垂体中AMA2的表达均与对照组差异不显著(P>0.05);试验Ⅲ组家兔丘脑-垂体-性腺轴和试验Ⅱ组家兔丘脑、垂体、睾丸中AMA1mRNA表达量均极显著低于对照组(P<0.01),试验Ⅱ组卵巢AMA1mRNA表达量显著低于对照组(P<0.05),试验Ⅲ组家兔睾丸和卵巢AMA2mRNA表达量显著低于对照组(P<0.05)。【结论】小花棘豆中毒可影响家兔丘脑-垂体-性腺轴AMA的活性及其基因的转录表达。【Objective】This study investigated the effects of Oxytropis glabra DC poisoning on content and transcriptional expression ofα-mannosidase(AMA)in hypothalamus-pituitary-gonad to reveal its toxicity mechanism.【Method】A total of 24 rabbits were randomly and equally divided into control group and experimental groupsⅠ,Ⅱ,andⅢ.The control group was feed with green hay only while the experimental groups were fed with mixed forage with 15%,30%and 45% dried O.glabra DC plant and the corresponding swainsonine contents were 30mg/kg,60mg/kg,and 90mg/kg,respectively.Two rabbits were random-ly selected from each group on the 14 th,35th and 70 th days to collect hypothalamus,pituitary and gonad for the measurement of distribution and content of AMA as well as the mRNA expression in different encephalic regions.【Result】AMA distributed in hypothalamus-pituitary-gonad in rabbits,and O.glabra DC reduced the distribution and content of hypothalamus-pituitary-gonad in experimental groups.AMA in golgi and lysosomal expressed in hypothalamus-pituitary-gonad of all experimental groups,and O.glabra DC reduced the expression of hypothalamus-pituitary-gonad AMA1 and AMA2mRNA.The difference in AMA1 and AMA2expressions was not significant between experimental groupⅠand control group in hypothalamus-pituitary-gonad at 70d(P〉0.05),so was the difference in AMA2 expression between experimental groupsⅡ andⅢ with control group.The difference in AMA1 expression was very significant between experimental groupⅢand control group in hypothalamus-pituitary-gonad at 70d(P〈0.01),so as the difference between experimental groupⅡ and control group in hypothalamus,pituitary and testis(P〈0.01).The difference was not significant between experimental group Ⅱ and control group in ovary(P〉0.05),but the difference of AMA2 expression was extremely significant between experimental groupⅢ and control group in testis and ovary(P〈0.01).【Conclusion】O.glabra DC had effects on content and transcriptional
