摘  要:目的:探索循经感传的物质基础。方法:SD大鼠60只,雌雄各半,随机分为常态组、常态针刺组、维拉帕米阻断组和0.9%NaCl溶液阻断组(对照组),每组15只。常态组及常态针刺组大鼠首先于足阳明胃经股骨段循经受观测部位(简称循经受观测部位)及邻近非经受观测部位(简称非经受观测部位)注射Fluo 3-AM(钙离子荧光探针)后孵育;维拉帕米阻断组先于循经受观测部位及非经受观测部位用维拉帕米注射液局部注射,再在循经和非经受观测部位注射Fluo3-AM后孵育;对照组用0.9%NaCl溶液局部注射,再在循经和非经受观测部位注射Fluo 3-AM后孵育。分别记录各组注射后的循经与非经受观测部位细胞内Ca^(2+)成像变化大于20min。后3组均在上述操作完成后,立即针刺"后三里",并连接电针1min,记录循经受观测部位和非经受观测部位细胞内Ca^(2+)成像变化大于20min。结果:常态组循经受观测部位细胞内Ca^(2+)荧光强度高于非经受观测部位;常态针刺组循经受观测部位针刺后细胞内Ca^(2+)荧光强度较针刺前明显增强,非经受观测部位针刺前后细胞内Ca^(2+)荧光强度未见明显变化;维拉帕米阻断组维拉帕米注射液局部阻断后,循经受观测部位细胞内Ca^(2+)荧光强度针刺后未见增强,非经受观测部位针刺前后细胞内Ca^(2+)荧光强度未见明显变化;对照组0.9%NaCl溶液局部注射后,循经受观测部位及非经受观测部位细胞内Ca^(2+)荧光强度未见明显变化,针刺前后细胞内Ca^(2+)荧光强度未见明显变化。结论:循经部位电压门控钙通道(voltage-gated calcium channels,VGCCs)状态与循经部位组织细胞兴奋传导具有高度相关性。Objective To explore the material basis of conduction along meridian.Methods Sixty SD rats(30males,30females)were randomly assigned into a normal group,an acupuncture group,a verapamil blocking group and a 0.9%NaCl blocking group(control group),15 rats in each one.Fluo 3-AM(calcium fluorescence probe)was injected at the observation part in femoral stomach meridian of foot-yangming(meridian part)and the approaching femoral meridian part(non-meridian part)in the normal group and the acupuncture group,and then incubation was applied.In the verapamil blocking group,verapamil was injected at local meridian part and non-meridian part,and in the control group 0.9%NaCl was injected.Then Fluo 3-AM was injected at the meridian part and non-meridian part in the two groups,and incubation was implemented.Ca^(2+)imaging changes in cells were recorded for more than 20 min after injection of every part in each group respectively.After the above operations in the last three groups,acupuncture was used at"Zusanli"(ST 36)immediately,with electroacupuncture for one min,then Ca^(2+)imaging changes in cells at the meridian and non-meridian parts were recorded for more than 20 min.Results In the normal group,Ca^(2+)fluorescence intensity at the meridian part was higher than that at the non-meridian part.In the acupuncture group,after acupuncture Ca^(2+)fluorescence intensity at the meridian part was obviously higherthan before,but the change before and after acupuncture was not apparent at the non-meridian part.After verapamil blocking local calcium channel and acupuncture,the Ca^(2+)fluorescence of the meridian part did not strengthen,and the change of that before and after acupuncture at the non-meridian part was not obvious.In the control group,after injecting 0.9% NaCl at local part,Ca^(2+)fluorescence intensities of the meridian and nonmeridian parts showed no obvious change,so was that before and after acupuncture.Conclusion The voltage-gated calcium channel at the m
