DA和SD大鼠在福尔马林诱发痛反应和机制的差异观察

摘  要:目的:观察福尔马林诱发的Dark-Agouti(DA)和Sprague Dawley(SD)大鼠痛反应差异。方法:分别采用行为学、电生理学、免疫组织化学方法观察福尔马林痛模型中DA和SD大鼠的伤害性行为、初级传入神经放电和脊髓背角磷酸化的c AMP反应元件结合蛋白(phosphorylated c AMP response element binding protein,p CREB)的表达。结果:DA和SD大鼠福尔马林注射后均出现典型的双相性伤害性行为,但是DA大鼠的抬舔爪行为更显著(P<0.01,P<0.001),SD大鼠的轻触时间和缩爪次数更显著(P<0.01,P<0.001)。福尔马林注射后1h内,DA和SD大鼠L5背根Aδ和C类纤维放电显著增加,以DA大鼠更显著(P<0.01)。福尔马林注射后DA和SD大鼠L4/L5脊髓背角Ⅰ-Ⅱ和Ⅴ-Ⅵ层p CREB表达均显著增加,但是SD大鼠更显著(P<0.001)。结论:结果提示DA和SD大鼠在福尔马林诱发的疼痛模型中存在不同的伤害性行为反应,并且其机制也不同。Objective: To observe the differences in nociceptive responses induced by formalin between Dark-Agouti( DA) and Sprague Dawley( SD) rats. Methods: Behavioral,electrophysiological and immunohistochemical methods were used to observe the nociceptive behaviors,primary nerve activities and expression of phosphorylated c AMP response element binding protein( p CREB) in dorsal horn of L4 /5 spinal cord in formalin pain model. Results: Formalin induced typical biphasic nociceptive behaviors in both DA and SD rats. However,the durations of lifting and licking were significantly higher in DA rats( P〈0. 01 and P〈0. 001),while the duration of favoring( P〈0. 001) as well as the number of flinches were higher in SD rats( P〈0. 01 and P〈0. 001). Consistent with lifting and licking,the Aδand C units were sensitized by formalin injection more in DA rats than those in SD rats( P〈0. 01). Formalin injection significantly increased the p CREB expression in both sides of laminae Ⅰ- Ⅱ and Ⅴ-Ⅵ in dorsal horn in both DA and SD rats( P〈0. 05). However,the pCREB expression was higher in SD rats compared with that in DA rats( P〈0. 001).Conclusion: These results suggest that there is significant individual difference in formalin pain model between the DA and SD rats,and the mechanisms underlying the various nociceptive behaviors are also different.
