摘 要:目的研究Wortmannin对糖尿病大鼠肺组织IL-1与IL-6表达的影响。方法 60只大鼠随机分为对照组(20)、糖尿病组(20)和Wortmannin处理组(20),通过高糖、高脂饮食加腹腔注射小剂量链脲佐菌素(STZ)的方法建立2型糖尿病大鼠模型。糖尿病模型建立后,检测血糖、血脂及体重的改变;采用免疫组织化学方法和Western blot方法检测各组大鼠肺组织中IL-1与IL-6表达的变化情况。结果糖尿病模型组大鼠血糖、血脂值比对照组明显增高(P<0.01),而Wortmannin处理组比糖尿病模型组明显降低(P<0.01)。免疫组织化学和Westernblot结果显示,糖尿病模型组大鼠肺组织Ⅱ-1与Ⅱ-6的表达量均显著高于对照组(P<0.01),而Wortmannin处理组比糖尿病模型组则显著降低。结论 PI3K/Akt信号通路可能通过调节Ⅱ-1与IL-6的表达参与糖尿病肺组织病变的发生发展。Objective To study the effect of Wortmannin on the expressions of IL-1 and IL-6 in lung tissue of diabetic rats. Methods 60 rats were divided into control group(20), diabetes mellitus group(20) and Wortmaunin treatment group(20). The diabetic rat models were established by giving high glucose, highfat diet combined with peritoneal injection with low-dose streptozotocin(STZ) . The values of blood glucose, blood lipids and body weight were measured after the establishment of the model of diabetes, the expressions of IL-1 and IL-6 were observed by immunohistochemistry and Western blot. Results The values of serum glucose and lipid were significantly higher in diabetes mellitus group than in the control group(p〈0.01), but lower significantly in Wortmannin treatment group than in diabetes mellitus group(P〈0.01). The expression levels of IL-1 and IL-6 in lung tissue were increased significantly in diabetes mellitus group than in control group(p〈0.01), but decreased significantly in Wortmannin treatment group than in diabetes mellitus group(P〈0.01). Conclusion PI3K/Akt signal pathway might be related to lung tissue pathological changes of dibtetic rats by regulating the expressions of IL- 1 and IL-6.
