基于叶绿体DNA rps16序列的福建泰宁野生铁皮石斛多样性分析

摘　　要：为了解福建泰宁野生铁皮石斛种源的多样性,对75份泰宁野生铁皮石斛及其他石斛属药用植物叶绿体DNArps16序列进行分析。结果表明:75份石斛属药用植物种间平均遗传距离为0.006 6,泰宁野生铁皮石斛种内平均遗传距离为0.00 06。38份泰宁野生铁皮石斛叶绿体DNArps16序列比对后,共得到5个多态性位点,多态性位点数占总数的25%,种内基于K-2P遗传距离最大为0.003 4,表明泰宁野生铁皮石斛叶绿体DNA rps16序列变异较大,多样性较高。此外,在泰宁野生铁皮石斛的5种rps16核苷酸序列中,T2型、T65型、D5型为参比铁皮石斛种质中的独有类型。A diversity analysis based upon the rps16 sequence of chloroplast DNA for the wild Dendrobium officinale found in Taining,Fujian and other areas was conducted.The results indicated an average genetic distance among 75 species of Dendrobiumto be 0.006 6,and 0.000 6for those from Taining.There were 5polymorphic sites（i.e.,25% of the total）detected in the 38 wild D.officinale from Taining.The maximal genetic distance calculated by K-2P was 0.003 4,indicating significant sequence variations and high diversities among the Taining species.In addition,among the 5different types of rps16 sequence,T2,T65,and D5 were found only in D.officinale collected from Taining.
