摘 要:目的:探讨人胶质瘤U251细胞的干细胞特性及定向诱导分化为脂肪细胞的潜能。方法:采用免疫荧光染色检测U251细胞中干细胞标志物巢蛋白,人类婆罗双树样基因4(sal-like 4,SALL4),神经细胞黏附分子(neural cell adhesion molecule,NCAM),胶质纤维酸性蛋白(glial fibrillary acidic protein,GFAP)的表达。采用成脂培养基诱导U251细胞,以CCK-8实验描记细胞生长曲线;油红-O染色及免疫印迹法检测脂肪细胞标志蛋白的表达。结果:胶质瘤细胞U251中巢蛋白,SALL4,NCAM,GFAP呈阳性表达;成脂诱导培养基定向诱导后,U-251细胞内大量橙红色脂滴形成;脂肪酸合酶、过氧化物酶体增殖物受体γ(PPARγ)、CCAAT增强子蛋白-β(CEBP-β)、脂联素表达增高。结论:人胶质瘤U251细胞具备干细胞表型,可定向诱导分化为脂肪细胞。Objective: To investigate the stem cell characteristics of human glioma cell line U251 and its potential of differentiating into adipocytes. Methods: Immunofluorescence staining was used to detect the expression of stem cell marker nestin, sal-like 4 (SALL4) , neural cell adhesion molecule ( NCAM ), glial fibrillary acidic protein (GFAP) in U251 cells. Growth curve of U251 cells after adipogenic induction was evaluated by CCK-8 method. After the induction, fat deposition was stained by Oil Red O staining and adi- pogenic marker expression was detected by Western blotting. Results: U251 cells showed immunofluores- cence positive staining of nestin, SALL4, NCAM, GFAP. After adipogenic induction, a large number of or- ange-red lipid droplets was seen in U-251 cells; Western blotting showed increased expression of adipocyte- specific proteins, such as fatty acid synthase, proliferator-activated receptor gamma(PPARγ) , CCAAT en- hancer-bingding protein-β(CEBP-β), adiponectin. Conclusion: U251 cells had stem cell phenotype and can be into adipocytes induction.
