摘 要:目的:观察酸浆水煎液对胃癌BGC-823细胞增殖的影响并探讨其机制。方法:将对数生长期的BGC-823细胞随机分为对照组、1.562 5、3.125、6.25、12.5、25 g/L实验组,分别用空白培养基和对应浓度酸浆水煎液培养24,48,72 h,倒置显微镜观察各组细胞生长状况和形态;MTT法检测各组细胞增殖活性;分别用3.125,6.25,12.5 g/L酸浆水煎液培养BGC-823细胞24,48 h,采用流式细胞术分析各组细胞周期。结果:对照组细胞排列紧密、形态规则,随酸浆水煎液浓度增加和作用时间延长,实验组BGC-823细胞逐渐出现接触松散,变圆变小,数量减少等生长抑制现象,12.5,25 g/L实验组变化明显;与对照组相比,不同浓度酸浆水煎液组细胞存活率明显降低(P<0.05);3.125,6.25,12.5 g/L酸浆水煎液作用细胞24,48 h后,G_2期细胞比例较对照组明显升高(P<0.05),且呈时间和浓度依赖性。结论:酸浆水煎液抑制胃癌BGC-823细胞增殖,且呈时间和浓度依赖性,其机制可能与细胞周期阻滞于G_2/M期有关。Objective: To study the effect of Physalis alkekengi L. decoction on the inhibition of human gastric cancer line BGC-823 cells proliferation, and to investigate its potential mechanism. Methods: The BGC-823 cells in logarithm growth phase were selected and randomly divided into control group and 1. 562 5, 3. 125, 6.25, 12.5, 25 g/L of Physalis alkekengi L. decoction groups. After BGC-823 cells were treated with DMEM and Physalis alkekengi L. decoction for 24, 48, 72 h respectively, MTT assay was used to detect the proliferation activities of BGC-823 ceils in various groups. The flow cytometry method was used to analyze the the cell cycle of BGC-823 cells treated with 0, 3. 125, 6.25, 12.5 g/L Physalis alkek- engi L. decoction for 24 and 48 h respectively. Results: After treated with Physalis alkekengi L. decoction, BGC-823 cells, especially in groups treated with 12.5, 25 g/L Physalis alkekengi L. decoction became loosely arranged, round and small and their numbers decreased compared with the control group. MTT as- say indicated that the cell viabilities in Physalis alkekengi L. groups were lower than those in control group ( P 〈 0.05 ). The percentage of cells in G2 phase in groups treated with Physalis alkekengi L. decoction at different concentration for 24 and 48 h were higher than that of control group ( P 〈 0.05 ). Conclusion : Physalis alkekengi L. decoction could inhibit the proliferation of BGC-823 cells in a time- and dose-depend- ent manner and its mechanism may relate to cell cycle G2/M arrest.
