摘 要:为有效控制枣疯病的发生,以‘狗头枣’和‘冬枣’为试材,研究了枣疯病对枣树离体器官再生的影响。结果表明,与健康组培苗相比,疯苗茎尖和茎段的腋芽更容易萌发,附加0.5mg/L IBA和1.0mg/L 6-BA可促进腋芽萌发。在MS+20g/L麦芽糖+5.5g/L琼脂+0.8mg/L 6-BA+0.2mg/L IBA培养基上,疯苗和健康苗叶片均有不定芽发生,且同一基因型内病苗和健康苗叶片出芽率差异不显著。在MS和1/2MS培养基上,疯苗和健康苗离体叶片均未发生不定根,但在附加4.0mg/L IBA之后疯苗离体叶片可以诱导得到不定根。在MS培养基中,疯苗和健康苗离体无根苗均可发生不定根,且疯苗生根率显著高于健康苗。附加0.2~1.0mg/L IBA后,疯苗无根苗生根率显著提高,而健康苗无根苗生根率无显著变化。发根农杆菌处理可显著促进狗头枣疯苗生根,但对狗头枣健康苗生根无显著影响。The effects of infection with jujube witches' broom disease(JWB)onin vitro organogenesis were studied using two cultivars of Ziziphus jujuba Mill.namely‘Goutouzao'and‘Dongzao'.Axillary buds at shoot tips and stems of tissue-cultured seedlings infected with JWB germinated more easily than healthy ones.Adventitious buds were formed from leaves of both infected and healthy seedlings in medium of MS+20g/L maltose+5.5g/L agar+0.8mg/L 6-BA+0.2mg/L IBA,and the rates of adventitious buds were not significantly different between the two types of tissue-cultured seedlings.Although no adventitious roots were obtained from leaves of both infected and healthy tissue-cultured seedlings in medium MS and 1/2 MS,the leaves of infected seedlings formed adventitious roots after adding supplement of 4.0mg/L IBA in the medium.Adventitious roots were formed in both two types of seedlings grown in the MS medium.The rate of adventitious roots was significantly higher in infected seedlings than in healthy ones.The rate of adventitious roots of infected seedlings was higherin 1/2MS+0.2-1.0mg/L IBA than in medium 1/2MS;however,the adventitious roots percentage in healthy seedlings had no significant difference in different media.In addition,the treatment with Agrobacterium rhizogenes significantly increased the rooting percentage in infected tissue-cultured seedlings,but not in healthy seedlings.
