摘 要:目的:通过观察荜茇根提取物对胆盐相关性胃炎模型大鼠抗肿瘤坏死因子-α(TNF-α)及环氧化酶-2(COX-2)的影响,探讨其治疗胆盐相关性胃炎的物质基础及作用机制。方法:140只SD大鼠分为14组:正常对照组;模型对照组;多潘立酮组(3.0mg·kg^(-1));乙醇提取物、乙酸乙酯提取物、正丁醇提取物高(400 mg·kg^(-1))、中(250 mg·kg^(-1))、低(125 mg·kg^(-1))剂量组;二氯甲烷提取物高(400 mg·kg^(-1))、低(125 mg·kg^(-1))剂量组,每组10只。观察各组大鼠胃窦组织病理组织学变化,放免法检测血清TNF-α浓度,RT-PCR法测定胃黏膜COX-2 mRNA的表达水平。结果:与模型组相比,荜茇根提取物各组胃窦组织炎症有不同程度减轻,血清TNF-α浓度及胃黏膜COX-2 mRNA表达水平也有不同程度降低,差异有统计学意义(P<0.05)。结论:荜茇根提取物(尤其是乙醇提取物)可减轻胆盐相关性胃炎模型大鼠胃黏膜炎症反应,其作用机制可能与抑制TNF-α及COX-2的表达有关。OBJECTIVE To discuss material basis and mechanism of bile salts correlation gastritis by observing effects of extract of Radix Piperis longi on TNF-αand COX-2in model rats with bile salts correlation gastritis.METHODS A total of 140 SD rats were divided into 14 groups:normal control group,model group,domperidone group(3.0mg·kg^-1),high dose(400mg·kg^-1),middle dose(250 mg·kg^-1)and low dose(125 mg·kg^-1)groups of Radix Piperis longi extract in ethanol,ethyl acetate and n-butyl alcohol,high dose(400 mg·kg^-1)and low dose(125 mg·kg^-1)groups of Radix Piperis longi extract in methylene dichloride,10 rats in each group.Pathological and histological changes of gastric antrum tissues of rats were observed,serum concentration of TNF-αwas determined by radioimmunoassay,COX-2 mRNA expression level of gastric mucosa measured by RT-PCR.RESULTS Compared with model group,extract of Radix Piperis longi reduced inflammation of gastric antrum tissues,declined serum concentration of TNF-αand gastric mucosa COX-2 mRNA expression level to different degrees,and the differences were statistically significant(P〈0.05).CONCLUSION Extract of Radix Piperis longi(especially in ethanol)can alleviate inflammatory response in bile salts correlation gastritis model rats,possibly through inhibiting expression of TNF-αand COX-2.
