白芷总香豆素对偏头痛模型大鼠PGE_2和TNFα的影响

摘　　要：目的:研究白芷总香豆素对偏头痛模型大鼠血中前列腺素E_2(PGE_2)及肿瘤坏死因子α(TNFα)的影响,探讨白芷总香豆素对治疗偏头痛的作用机理。方法:采用SD大鼠皮下注射硝酸甘油复制大鼠偏头痛模型,实验组灌胃给予不同剂量白芷总香豆素混悬液,给药4 h后,将动物断头取血,分离血清,采用酶联免疫双抗体夹心法检测血清中PGE_2和TNFα含量。结果:白芷总香豆素在0.72 g.kg^(-1)剂量下可明显抑制模型动物血清中PGE_2水平,降低模型动物血中TNFα含量,与模型组比较有明显差异(P<0.05)。结论:白芷总香豆素可通过抑制TNFα、PGE_2释放来发挥抗炎镇痛作用,可能为其临床治疗偏头痛作用机理之一。Objective： To study the effects of Baizhi total coumarin on PGE2 and TNFct in rat migraine model, and investigate the mechanism of Baizhi total coumarin in treating migraine. Method： The rat migraine model was induced by hypodermic injection of nitroglycerinum and given different dosages of Baizhi total coumarin. The serum of rat migraine model was collected to detect the levels of PGE2 and TNFct by ELISA after 4h. Result： The levels of PGE2 and TNFct were obvious lower in the serum of rat migraine model administered with 0.72 g. kg-1 Baizhi total coumarin compared with the control group （P 〈0. 05）. Conclusion： Baizhi total coumarin shows analgesic and anti-inflammatory effects through restraining TNFct and PGE2, which maybe one of mechanisms for Baizhi total coumarin in treating migraine in clinic.
