摘 要:目的:观察护骨胶囊对去卵巢大鼠腰椎松质骨骨组织形态计量学的影响。方法:3月龄雌性SD大鼠60只,随机分为假手术(Sham)、去卵巢(OVX)、仙灵骨葆胶囊阳性对照(Pos)和护骨胶囊低(HG-L)、中(HG-M)、高(HG-H)剂量组,每组10只。Sham组仅去除卵巢周围部分脂肪,其他组均去除卵巢。术后2 d开始给药,持续3个月后,采用骨组织形态计量学检测腰椎骨的动静态参数。结果:与Sham组比较,OVX组骨小梁面积百分数(%Tb.Ar)、骨小梁数量(Tb.N)降低、骨小梁分离度(Tb.Sp)增加(P<0.01),单位骨小梁周长成骨细胞(Ob.N)、成骨细胞周长百分数(%Ob.Pm)和单位骨小梁周长破骨细胞数(Oc.N)、破骨细胞周长百分数(%Oc.Pm)增加(P<0.01或P<0.05)。与OVX组比较,HG-L、HG-M、Pos组%Tb.Ar均升高(P<0.01或P<0.05),HG-L组Tb.N升高(P<0.01),而HG-L、HG-M和Pos组Tb.Sp均降低(P<0.01或P<0.05),HG-L组荧光周长百分数(%L.Pm)、骨形成率BFR/BV和BFR/BS均降低(P<0.01或P<0.05),HG各剂量组和Pos组%Ob.Pm均增加(P<0.01或P<0.05),Pos组Oc.N降低(P<0.05)。结论:护骨胶囊可以有效地增加去卵巢大鼠腰椎松质骨的骨量、改善骨微结构,低剂量抑制骨高转换,且促进骨形成,与仙灵骨葆比较,改善骨结构效果明显。AIM: To study the effect of Hugu capsules( HG) on bone histomorphometry parameters of lumbar cancellous bone in ovariectomized( OVX)rats. METHODS: Sixty female 3-month-old SD rats were randomly divided into Sham,OVX,Pos,HGL,HG-M,and HG-H groups,10 in each group. All rats were ovariectomized while rats in Sham group were only removed of the fat around the bilateral ovaries. At the 2nd day after the operation,all the rats were given medicine for three months and then killed. The dynamic and static parameters of lumbar vertebrae were tested by bone morphometry. RESULTS: Compared to Sham group,Tb. Sp,Ob. N,% Ob. Pm,Oc. N,% Oc. Pm increased( P〈0. 01 or P〈0. 05),but % Tb. Ar and Tb. N decreased( P〈0. 01) in OVX group. Compared to OVX group,%L. Pm,BFR / BV and BFR / BS decreased in HG-L group. % Tb. Ar increased while Tb. Sp decreased in HG-L,HG-M and Pos groups( P〈0. 01 or P〈0. 05),Tb. N also increased in HG-L group( P〈0. 01). % Ob. Pm increased in all HG and Pos groups( P〈0. 05 or P〈0. 01) and Oc. N decreased in Pos group( P〈0. 05). CONCLUSION: HG capsules can better improve bone histomorphometry indexes of lumbar in OVX rats than Xianlinggubao Bao,not only by increasing the cancellous bone mass,improving bone microstructure and promoting bone formation,but also by inhibiting the high bone turnover state.
