摘  要:目的探讨血管内皮生长因子(vascular endothelial growth factor,VEGF)和基质金属蛋白酶-9(matrix metalloproteinase-9,MMP-9)与子痫前期发病的相关性。方法纳入河北省沧州市中心医院2013年1月至2014年11月确诊为子痫前期的患者100例,其中轻度子痫前期的患者51例,重度子痫前期患者49例,同时纳入同期分娩的健康孕妇50例作为对照组。采用Western blotting、反转录酶-聚合酶链反应分子生物学方法检测3组患者胎盘组织中VEGF、MMP-9的表达水平;采用酶联免疫吸附方法检查患者血清及尿液中VEGF、MMP-9的水平。结果轻、重度子痫前期组患者的胎盘组织中VEGF、MMP-9的表达水平均低于正常对照组,且组间相比差异有统计学意义(P<0.05);轻、重度子痫前期组患者血清及尿液中VEGF、MMP-9的水平均低于正常对照组,且组间相比差异有统计学意义(P<0.05)。结论 VEGF、MMP-9参与了子痫前期的病理生理改变,且与病情的发生及轻重程度有密切相关性。Objective To investigate the correlation of vascular endothelial growth factor (VEGF) , matrix metalloproteinase-9 (MMP- 9) and incidence of preeclampsia. Methods 100 patients diagnosed with preeclampsia in Cangzhou Central hospital from January 2013 to November 2014 were selected, including 51 cases of mild pre-eclampsia and 49 cases severe preeclampsia, while 50 healthy pregnant women during the same period were set as the control group. Using Western blotting, RT - PCR molecular biology methods to detect VEGF and MMP - 9 expression levels in patients' plaeent of three groups ; using ELISA method to check VEGF and MMP -9 serum levels of serum and urine. Results The expression levels of VEGF and MMP -9 in placental tissue in patients with preeclampsia were lower than those in the control group, with a statistically significant difference ( P 〈 0.05 ) ; the levels of VEGF and MMP -9 in serum and urine in patients with preeclampsia were lower than those in the control group, with a significant difference (P 〈 O. 05). Conclusion VEGF and MMP - 9 involve in the pathophysiology of preeclampsia, and have a close correlation with the occurrence and severity of the disease.
