规律运动对心脏特异性Ca-α1D基因沉默老龄果蝇心脏节律和收缩功能的影响

摘  要:目的:研究青年期开始规律运动对心脏Ca-α1D基因沉默老龄果蝇心脏功能、运动能力和生命周期的影响,探讨Ca-α1D基因在心脏衰老及运动抗心脏衰老中的作用。方法:基因型为w1118;P{GD1737}v51491的处女蝇分别与W1118品系、Hand-GAL4品系雄蝇杂交,获得F1代处女蝇分为安静组、干扰组、运动组、运动干扰组,每组400只,饲养至2周龄开始运动,2小时/天,5天/周,训练持续时间为4周。采用qRT-PCR测定心肌Ca-α1D基因mRNA表达量,M-mode心动图检测评价心脏功能,测量攀爬指数评定运动能力,统计生命周期反映寿命情况。结果:(1)干扰组心肌Ca-α1D的相对表达量较对照组约下降50%(P<0.01)。(2)与对照组相比,干扰组心动周期(HP)增加(P<0.01),收缩间期(SI)上升(P<0.05)。纤维性震颤(FL)、心动徐缓发生率增加(P<0.01)。舒张直径(DD)、收缩直径(SD)增加(P<0.01),射血分数(FS)降低(P<0.05)。与对照组相比,运动组HP、DI上升(P<0.05),心动徐缓发生率增加(P<0.01)。与对照组相比,运动干扰组HP、心动徐缓发生率增加(P<0.01)。DD、SD增加(P<0.01),而FS变化不具有显著性差异(P>0.05)。与干扰组相比,运动干扰组心动徐缓发生率减少(P<0.01)。DD增加(P<0.05)。基因沉默联合耐力运动对心动徐缓发生率的结果有协同作用(P<0.05)。其中基因是对HP、DD、SD、FS、FL结果产生作用的主效应,运动是对DI结果产生作用的主效应。(3)与对照组相比,运动组攀爬能力增强(P<0.01)、干扰组攀爬能力降低(P<0.05);基因沉默联合耐力运动对攀爬能力具有协同作用(P<0.05)。(4)运动组平均寿命高于对照组(P<0.01),运动干扰组高于干扰组(P<0.01),干扰组平均寿命低于对照组(P<0.01)。结论:规律运动的干预能够维持心脏正常节律、提高运动能力和增进生命周期有一定作用。心脏特异性Ca-α1D基因沉默是导致该果蝇心脏收缩功能减退、节律紊乱以及寿命缩减的主要原因。The objective of this paper was to study the effects of regular exercise on the cardiac function,exercise capacity and life cycle of Drosophila with cardiac Ca-α1D gene silence at adolescence,and to explore the function of Ca-α1Dand training on cardiac aging.The virgin female flies with w1118;P{GD1737}v51491genotype were crossed with W1118 flies and Hand-Gal4 flies respectively,then the virgin female F1 progeny flies were respectively divided into control groups,interference group,exercise groups and interference group of exercise.The flies in the exercisegroups at 15 days old performed platform 2hours per day for,5days a week for 4weeks.The Ca-α1DmRNA expression in cardiac muscle was measured by qRT-PCR and heart function was recorded by M-mode echocardiography.The exercise capacity was assessed by climbing index and lifespan was reflected by lifecycle.The results revealed that:(1)The expression of Ca-α1Din RNA interference group was approximately decreased by 50% compared to control group(P〈0.01).(2)Compared with the control group,the interference group systolic intervals(SI)increased(P〈0.05),heart period(HP),Fibrillations(FL),bradycardia rate increased(P〈0.01),diastolic diameter(DD),systolic diameter(SD)increased(P〈0.01),fractional shortening(FS)reduced(P〈0.05).Compared with the control group,the interference group of exercise HP,bradycardia rate increased(P〈0.01).SD,DD increased(P〈0.01),FS had not significant change(P〈0.05).And compared to the control group,exercise group HP,DI increased(P〈0.05),bradycardia rate increased(P〈0.01).Compared with interference group,the interference group of exercise bradycardia rate reduced(P〈0.01),DD rise(P〈0.05).Gene silencing and endurance exercise combined had a synergistic effect on HR(P〈0.05).The primary effect of HP,DD,SD,FS,FL was gene.Exercise was the main effect of DI resulting effects.(3)Compared with the control group,the exercise group of climbing
