摘  要:目的建立测定夏天无滴眼液中原阿片碱含量的HPLC法。方法采用ZORBAX SB-C18(150 mm×46 mm,5μm)色谱柱,以乙腈-0.1%三氟乙酸溶液(25∶75)为流动相,柱温30℃,流速1.0 m L/min,检测波长290 nm,进样量20μL。结果在该色谱条件下,原阿片碱在浓度14.2-142μg/m L范围内线性关系良好(r=0.9999,n=9),加样回收率为104.5%,RSD为2.47%,日内和日间精密度RSD均小于1%(n=5),样品在24 h内稳定。结论本方法简单,结果准确可靠,重复性好,可以用于测定夏天无滴眼液中原阿片碱的含量。Objective To establish an HPLC method for determination of content of protopine in rhizoma corydalis decumbentis eye drops.Methods The HPLC separation was carried out on a ZORBAX SB-C18(150 mm × 46 mm,5 μm) column,the mobile phase was acetonitrile-0. 1% TFA solution(25∶75) with the velocity of 1 m L/min and the column temperature of 30 ℃,the detection wavelength was set at 290 nm. And the injection volume was 20 μL. Results The good linearity of protopine under chromatography was found of 14. 2-142 μg/m L(r = 0. 9999,n = 9),The average recovery rate was 104. 5% with RSD of 2. 47%,Both intra-and inter-day precision RSD were less than 1%(n = 5). The samples were stable in 24 h.Conclusion The method is simple,accurate and repeatable,it can be used for the quality control of protopine in rhizoma corydalis decumbentis eye drops.
