艾菲提蒙汤对动脉粥样硬化形成家兔脂质过氧化和血管内皮功能的影响

摘　　要：目的：探讨艾菲提蒙汤在家兔实验性动脉粥样硬化（AS）模型形成过程中的抗脂质过氧化和血管内皮保护作用。方法：48只家兔随机分为6组,正常组、模型组、阳性对照组（舒降之）和艾菲提蒙汤高、中、低剂量组,每组8只。采用高脂饲料喂养60 d,建立AS家兔模型。同时各组家兔灌胃给予相应药物及生理盐水,60 d后测定各组家兔血清SOD、CAT、GSH-Px活力及NO、MDA含量,并采用荧光定量PCR技术检家兔主动脉内皮型e NOS、LOX-1及肝脏LDL-R表达。结果：艾菲提蒙汤在3.3~13.0 g/kg剂量范围内,可明显升高AS家兔血清中SOD、CAT、GSH-Px活性和NO含量,降低MDA含量,降低家兔主动脉内皮型e NOS、LOX-1和肝脏LDL-R的表达,与模型组比较差异均具有统计学意义（P〈0.05~0.01）。结论：艾菲提蒙汤可通过抑制AS家兔抗脂质过氧化、保护血管内皮功能而预防AS的发生。Objective： To explore the effects of Aifei Timeng Tang on lipid peroxidation and endodermis of blood vessel during the formation process of experimental atherosclerosis in rabbits. Methods： 48 rabbits were divided into 6 groups,normal,model,positive groups and low,middle,high dose Aifei Timeng Tang groups,with each 8 rabbits. Atherosclerosis models of rabbits were built with feeding high- fat diet for 60 days,each medicine group was given the corresponding medicine for 60 days at the same time,then rabbits＇ serum SOD,CAT,GSH- Px,NO,MDA were determined,and the expressions of e NOS,LOX- 1,LDL- R in rabbits＇ aorta endothelium and liver were detected by fluorescent quantitative PCR technology. Results： Aifei Timengi Tang could raise serum SOD,CAT,GSH- Px,NO / evels and lower MDA contents,inhibit the expressions of e NOS,LOX-1,LDL- R in rabbits＇ aorta endothelium and liver at the dose 3. 3 ~ 13. 0 g / kg,the differences were significant compared with model group rabbits. Conclusion： Aifei Timeng Tang can inhibit the formation of rabbits＇ experimental AS,inhibit the lipid peroxidation and protect blood vessel endothelium.
