几种佐剂对H7N9流感病毒灭活疫苗免疫增强效果的实验研究

摘　　要：目的为加强流感病毒H7N9灭活疫苗的免疫效果,本研究以BALB/c小鼠为模型,比较了Al(OH)3、poly(I∶C)、CpG、PEI四种佐剂对H7N9全病毒灭活疫苗和裂解疫苗的免疫增强效果。方法将四种佐剂分别与同剂量的流感病毒H7N9全病毒灭活疫苗或裂解疫苗联合免疫,在肌内注射免疫3周后使用40×LD50的同源病毒攻击小鼠,通过检测小鼠免疫后的抗体水平及攻毒后的小鼠存活率、体质量丢失情况及肺部病毒滴度来评价疫苗联合佐剂的免疫效果。结果四种佐剂均能提高两种灭活疫苗的抗体水平,此外,裂解疫苗和全病毒疫苗单独免疫时小鼠的存活率分别为33.3%、90.0%。除poly(I∶C)对全病毒苗没有显示出增强保护力的佐剂效果外,其他添加佐剂组小鼠均可达100.0%存活。结论全病毒疫苗+Al(OH)3和裂解疫苗+CpG诱导的免疫效果最佳,表明佐剂效应的发挥,会受到疫苗类型的影响。Objective To improve the immune protection provided by inactivated H7N9 influenza virus vaccine,four kinds of adjuvants,including Al（OH）3,poly（I∶C）,CpG and PEI,were added,respectively,to the H7N9whole-virion vaccine and split vaccine,and their adjuvant effects were compared in BALB/c mouse model. Methods Mice were immunized intramuscularly once with whole-virion or split inactivated H7N9 virus vaccine at the same HA dosage alone or with one of the four adjuvants.Three weeks after the immunization,the mice were challenged with a lethal dose（40×LD50）of the homologous virus.The protective effects of the vaccinations were evaluated by detecting vaccine specific antibody titer in serum,survival rate,lung viral titer and body weight loss of mice. Results The results showed that all the four adjuvants could increase the levels of specific antibody response induced by the vaccines.The survival rates of mice immunized by the split vaccine and whole-virion vaccine alone were 33.3% and 90.0%,respectively.Except for the whole-virion vaccine＋poly（I∶C）combination,all other combinations provided 100.0% protection against challenge. Conclusions The whole-virion vaccine＋Al（OH）3and split vaccine＋CpG were the two best combinations,which demonstrated that the effect of an adjuvant was influenced by the type of vaccine to some extent.
