摘  要:目的探讨腺病毒介导的肿瘤生长抑制因子4(Ad-ING4)在体外对人胃癌AGS细胞凋亡的作用及其p53基因表达的影响。方法不同浓度(80、160、320ng/ml)的Ad-ING4作用人胃癌AGS细胞48h后,用Annexin V-FITC检测细胞凋亡率,RT-PCR检测p53 m RNA的表达,免疫印迹法检测p53蛋白表达水平,并与对照组进行比较。结果凋亡检测发现,不同浓度Ad-ING4可明显诱导人胃癌AGS细胞凋亡,并呈浓度依赖性(P<0.05);Ad-ING4作用前AGS细胞p53 m RNA和p53蛋白表达水平较低,不同浓度Ad-ING4作用后p53 m RNA和p53蛋白表达水平明显上调,呈浓度依赖性(P<0.05)。结论随Ad-ING4作用浓度增高,人胃癌AGS细胞出现明显凋亡,其机制可能与p53基因转录活性增强有关。Objective To investigate the effects of adenovirus-mediated ING4 (Ad-ING4) on apoptosis and expression of p53 gene in human gastric AGS cells. Methods AGS cells were treated with Ad-ING4 at different concentrations (80, 160,320ng/ml). Cell apoptosis was evaluated by AnnexinV-FITC/PI fluos stainging. Results AnnexinV-FITC/PI showed that the apoptosis rate in AGS cell went up significantly after treatment of Ad-ING4 in a dose dependent manner (P〈0.05). Compare to control group, the mRNA and protein expressions of p53 gene were increased in a dose dependent manner after treaded with Ad-ING4 at different concentrations (P〈0.05). Conclusion Ad-ING4 can induce apoptosis of gastric cancer AGS cells, which may be related to the enhancement of the transcriptional activity of p53 gene.
