摘  要:目的:探讨神经鞘内转染p300si RNA(si RNA-p300)对大鼠神经病理痛阈值的影响。方法:成年雄性清洁级SD大鼠27只随机分为假手术组、对照组和实验组,每组9只。对照组和实验组应用坐骨神经压迫损伤建立神经病理性痛模型并鞘内置管,分别鞘内转染无关4μg si RNA和4μg si RNA-p300。假手术组单纯进行模拟手术并注射等量生理盐水。分别于转染前及注射后2、4、6、8、10、12和14 d分别检测3组大鼠痛阈分值,于注射后14 d处死大鼠,取腰段脊髓采用Western-blot法检测大鼠脊髓中p300表达水平。结果:与假手术组(21.22±1.56)比较,对照组(14.33±4.02)和实验组(13.66±2.30)大鼠疼痛阈值在转染后第2 d开始显著降低(P<0.05)。与对照组(8.98±2.89)比较,实验组(11.23±2.20)大鼠疼痛阈值在转染后第6天开始显著升高(P<0.05);假手术组、对照组和试验组p300相对表达量分别为0.11±0.03、0.67±0.18和0.22±0.04,与假手术组比较,对照组和实验组p300表达显著上调(P<0.05);与对照组比较,实验组p300表达明显下调(P<0.05)。结论:鞘内注射si RNA-p300可显著降低大鼠神经病理性疼痛,提高痛阈。Objective tion of si RNA-p300 in neuropathic pain rats.Methods Twenty-seven adult male SD rats were randomly divided into sham operation group, control group and si RNA-p300 group with 9 in each group. Rats in the control group and si RNA-p300 group were induced by chronic constrictive injury and intrathecal independent si RNA 4μg and si RNA-p300 4 μg in the control group and si RNA-p300 group respectively. Rats in the sham operated group were intrathecal injection of 0.9% normal saline. The pain score were recorded and compared among the three groups at the time point of 2,4,6,8,10,12 and 14 days. After 14 days of given si RNA-p300, the rats were executed and the p300 protein were tested by western blot assay.Results Compared with the sham operated group(21.22±1.56), the pain threshold in the control(14.33±4.02) and si RNA-p300 group(13.66±2.30) were significant decreased in day 2(P〈0.05). Compared with control group(8.98 ± 2.89), the pain threshold in si RNA-p300 group(11.23±2.20) were significant elevated in day 6(P〈0.05); The p300 relative expression were0.11±0.03、0.67±0.18 and 0.22±0.04 in sham operated group, control group and si RNA-p300 group. Compared with shamed operated group, the p300 expression was significant evevated in the control and si RNA-p300 group(P〈0.05). Compared with control group, the p300 expression in the si RNA-p300 group was statistical decreased(P〈0.05).Conclusion The neuropathic pain can be decreased by intrathecal si RNA-p300 in neuropathic pain rats.
