摘 要:目的:建立HPLC同时测定败酱草中齐墩果酸、熊果酸和常春藤皂苷元3种三萜类成分的"一测多评"法。方法:以齐墩果酸为内参物,采用HPLC测定含量,建立其与熊果酸和常春藤皂苷元的相对校正因子(RCF),以相对保留时间(RRT)进行色谱峰定位,计算熊果酸和常春藤皂苷元的量;用外标法测定6批次败酱草中熊果酸和常春藤皂苷元的量,比较计算值与实测值的差异,验证一测多评的可行性和重现性。结果:测得熊果酸和常春藤皂苷元的平均相对校正因子为0.860(RSD1.24%)、0.934(RSD 0.79%);在不同HPLC和色谱柱下,熊果酸和常春藤皂苷元RCF的RSD分别为0.85%、0.72%,RRT的RSD分别为0.71%、1.07%;熊果酸和常春藤皂苷元的计算值和实测值之间的夹角余弦值分别为0.999 97和0.999 89,两种方法所测得的两种成分的含量相对偏差都在±5%以内。结论:一测多评法用于败酱草的三萜类多成分含量测定准确可行,重现性好。Objective To establish a quantitative analysis method of multi-components by single-marker(QAMS) for determinating oleanolic acid,ursolic acid and hederagenin simultaneously in Herba Patriniae by HPLC.Methods The content of internal reference substance Oleanolic acid was determined by HPLC, and the relative correlation factors(RCF) of oleanolic acid and ursolic acid, oleanolic acid and hederagenin were established. The chromatographic peaks of ursolic acid and hederagenin were located by the relative retention time(RRT). The contents of the two determinands were calculated according to the respective RCF and the injection content of oleanolic acid. The contents of ursolic acid and hederagenin in 6 batches Herba Patriniae were determined in the external standard method and were contrasted with the calculated value to verify the feasibility and repeatability of the QAMS method.Results The RCF of ursolic acid and hederagenin with reference to Oleanolic acid were 0.860(RSD 1.24%) and 0.934(RSD 0.79%)respectively. In different HPLC and column, the RSD of the RCF were 0.85% and 0.72% respectively, and the RSD of the RRT were 0.71% and 1.07% respectively. The included angle cosine value between the calculated and the measured value of ursolic acid and hederagenin were 0.999 97 and 0.999 89 respectively, and the relative deviation of the contents in two methods all were less than ±5%.Conclusion The QAMS is a kind of accurate and feasible method to determine the contents of the multi-components simultaneously in Herba Patriniae.
