摘  要:目的研究姜黄素对人肝癌Bel-7402细胞的抑制作用,并探讨其作用机制。方法实验分为4组,分别为对照组和10、20和40μmol/L姜黄素组。采用噻唑蓝(MTT)法检测细胞的活性;流式细胞术结合Annexin V-FITC/PI双染检测Bel-7402细胞的凋亡情况;RT-PCR法检测Bax和Bcl-2的mRNA的表达,Western blot检测细胞色素C和Caspase-3蛋白的表达。结果姜黄素显著抑制Bel-7402细胞的活性(P<0.01)。姜黄素处理后,Bel-7402细胞的凋亡率明显增加,与对照组相比具有统计学差异(P<0.05),且具有剂量依赖性。另外,姜黄素使Bax的mRNA表达上升,Bcl-2的mRNA表达下降,Bax/Bcl-2比值明显上升(P<0.05)。同时,细胞色素C和Caspase-3蛋白的表达均显著上调(P<0.01)。结论姜黄素可以抑制Bel-7402细胞的活性和增殖,促进Bel-7402细胞的凋亡,其机制可能与与姜黄素激活Bel-7402细胞线粒体凋亡途径有关。Objective To research the inhibitory effects of curcumin on human hepatoma carcinoma Bel-7402 cells and to investigate its possible mechanisms.Methods Experiments were divided into four groups,including control group,and 10,20 and 40μmol/L curcumin-treated groups.Cell activity of Bel-7402 was detected by MTT after Bel-7402 cells treated with different concentrations of curcumin.The rate of Bel-7402 apoptosis was tested by flow cytometry binding double staining assay of Annexin V-FITC/PI.RT-PCR was used to detect the mRNA expression of Bax and Bcl-2,and then the protein expression of cytochrome C and Caspase-3also were measured by Western blotting.Results Curcumin significantly inhibited the activity and growth of Bel-7402cells(P〈0.01).After curcumin treatment,the apoptosis rate of Bel-7402 cells was obviously increased compared with control group(P〈0.05),and showed a dose-dependent relationship.Moreover,the mRNA expression of Bax was increased by curcumin-treatment,inversely,Bcl-2was decreased,and the ratio of Bax/Bcl-2was significantly increased(P〈0.05).Meanwhile,the protein expression of cytochrome C and Caspase-3were also dramaticlly upregulated by curcumin-treatment(P〈0.01).Conclusion Curcumin can inhibit the activity and proliferation of Bel-7402 cells,and promote apoptosis process of Bel-7402 cells,which may be related with curcumin activate mitochondrial apoptosis pathway in Bel-7402 cells.
