摘  要:目的观察补阳还五汤对L-蛋氨酸(L-met)致高脂血症大鼠模型血脂的调节作用。方法采用SD大鼠灌胃L-蛋氨酸溶液复制高脂血症大鼠模型,造模同时给予阿托伐他汀(0.01g/kg)和补阳还五汤(5、10、20g/kg)干预,连续8周;在给药前及给药后2、4、8周采血,检测各组大鼠血清甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)及高密度脂蛋白胆固醇(HDL-C)水平并计算动脉粥样硬化指数(AI)。结果与正常组比较,L-蛋氨酸灌胃后,模型组大鼠血清TC[2周:(1.78±0.27)mmol/L,4周:(1.47±0.13)mmol/L,8周:(1.59±0.12)mmol/L]、TG[2周:(0.76±0.10)mmol/L,4周:(0.78±0.15)mmol/L,8周:(0.75±0.05)mmol/L]、LDL-C([2周:(0.29±0.06)mmol/L,4周:(0.25±0.03)mmol/L,8周:(0.22±0.04)mmol/L]水平明显增高(P<0.05,P<0.01),动脉粥样硬化指数(AI)(2周:1.42±0.32,4周:1.23±0.37,8周:1.37±0.12)显著增大(P<0.01);与模型组比较,补阳还五汤10、20g剂量能显著降低模型组大鼠血清TC[10g:2周:(1.48±0.22)mmol/L,4周:(1.21±0.14)mmol/L,8周:(1.35±0.08)mmol/L;20g:2周:(1.53±0.18)mmol/L,4周:(1.28±0.09)mmol/L,8周:(1.43±0.13)mmol/L]、TG([10g:2周:(0.63±0.07)mmol/L,4周:(0.60±0.09)mmol/L,8周:(0.69±0.08)mmol/L;20g:4周:(0.63±0.15)mmol/L]、LDL-C[10g:2周:(0.18±0.05)mmol/L,4周:(0.20±0.03)mmol/L,8周:(0.16±0.03)mmol/L;20g:2周:(0.23±0.09)mmol/L,4周:(0.19±0.04mmol/L,8周:(0.17±0.03)mmol/L]水平及AI(10g:2周:0.97±0.19,4周:0.94±0.12,8周:0.88±0.29;20g:2周:1.06±0.27,4周:0.89±0.13,8周:1.00±0.25;P<0.05,P<0.01)。结论补阳还五汤对L-met致异常的大鼠血脂水平具有良好的调节作用,对大鼠动脉粥样硬化具有一定的防治作用。Objective To investigate the effect of Buyang huangwu decoction(BYHWD) on blood lipids levels in rats with hyperlipidaemia induced by L-methionine. Methods Hyperlipidaemia model rats were made by ig. Lmethionine for 8 weeks on end, atorvastatin(0.01 g/kg) and BYHWD(5, 10, 20 g/kg) was administrated at the same time. At 2, 4, and 8 weeks, blood samples were collected from venous sinus of eye-orbit in rats, and total cholesterol (TC), triglycerides (TG), low-density lipoprotein-cholesterol (LDL-C), high-density lipoprotein-cholesterol (HDL-C) levels in sera were measured by fully automatic biochemical analyzer according to the kits instruction and atherosclerosis index (AI) was calculated. Results Compared with control group, the serum levels of TC, TG and LDL-C and AI in model rats at 2, 4, and 8 weeks were significantly increased(TC: 1.78 0.27 mmol/L, 1.47 0.13 mmol/L, 1.59 0.12 mmol/L; TG: 0.76 0.10 mmol/L, 0.78 0.15 mmol/L, 0.75 0.05 mmol/L; LDL-C: 0.29±0.06 mmol/L, 0.25±0.03 mmol/L, 0.22±0.04 mmol/L; P〈0.05 or P〈0.01; Ah 1.42±0.32, 1.23±0.37, 1.37±0.12, P〈0.01). Compared with model group, BYHWT with dosage of 10, 20 g/kg significantly lowered blood lipid levels and AI at 2, 4, and 8 weeks (TC with dosage of 10 g/kg: 1.48±0.22 mmol/L, 1.21±0.14 mmol/L, 1.35±0.08 mmol/L;TC with dosage of 20 g/kg: 1.53±0.18 mmool/L, 1.28±0.09 mmol/L, 1.43±0.13 mmool/L; TG with dosage of 10 g/kg: 0.63±0.07 mmool/L, 0.60±0.09 mmol/L, 0.69±0.08 mmool/L; TG with dosage of 20 g/kg: 0.63±0.15 mmool/L; LDL-C with dosage of 10 g/kg: 0.18±0.05 mmool/L, 0.20±0.03 mmool/L, 0.16±0.03 mmol/L; LDL-C with dosage of 20 g/kg: 0.23±0.09 mmol/L, 0.19±0.04 mmol/L, 0.17±0.03 mmool/L; AI with dosage of 10 g/kg: 0.97±0.19, 0.94±0.12, 0.88±0.29; AI with dosage of 20 g/kg: 1.06±0.27, 0.89±0.13, 1.00±0.25; P〈0.05 or P〈0.01).
