摘 要:目的观察定喘汤及其宣、降、清分解剂对RSV感染大鼠T细胞亚群及NK细胞活性的影响,探讨定喘汤全方及其"宣降清"三法的细胞免疫调节作用。方法将Wistar大鼠随机分为正常组、模型组、定喘汤组、宣法组、降法组及清法组6组,每组各9只,动物造模后正常组和模型组均以等体积的生理盐水灌胃,各干预治疗组分别予相应的中药灌胃。采用流式细胞术检测各组大鼠外周血中T淋巴细胞亚群及NK细胞活性。结果与正常组比较,模型组的CD8+细胞、CD3-CD161+细胞的百分比均下降,CD4+/CD8+比值升高具有显著性差异(P<0.05);定喘汤组和清法组的CD4+/CD8+比值下降与模型组比较具有显著性差异(P<0.05),定喘汤组和宣法组的CD3-CD161+细胞百分比升高与模型组比较也具有统计学意义(P<0.05)。结论定喘汤及其宣、清分解剂均对呼吸道合胞病毒感染Wistar大鼠后的免疫功能紊乱具有一定的调节作用。Objective In order to evaluate the immunoregulatory effects of asthma-relieving decoction and its ascending( xuan)therapy、descending( jiang) therapy、pyretic clearing( qing) therapy,the variety of T cell subset and NK cell were observed in rats with respiratory syncytial virus with dingchuantang. Methods Wistar rats were equally divided into 6 groups randomly: normal group、model group、asthma-relieving decoction group、ascending( xuan) therapy group、descending( jiang) therapy group、pyretic clearing( qing) therapy group,9 rats in each group. After animal models of respiratory syncytial virus infected rats were established successfully,each treatment group was intragastric administration daily with drug respectively. The normal group and model group were given nomal saline by gavage daily. Then the variety of T cell subset and NK cell were measured with flow cytometry. Results Compared with the normal group,the falled level of CD8+cells and CD3-CD161+cells were significantly different as same as the increasing rate of CD4+/ CD8+in the model group( P〈0. 05). The ratio of CD4+/ CD8+were lower in asthma-relieving decoction group and pyretic clearing( qing) therapy group than that of the model group significantly( P〈0. 05). The variation of CD3-CD161+cells were also statistically significant in asthma-relieving decoction group and ascending( xuan) therapy group( P〈0. 05). Conclusion Respiratory syncytial virus led to immune function disorder in Wistar rats,and the asthma-relieving decoction group、ascending( xuan) therapy group、pyretic clearing( qing) therapy group could effectively regulate immune function.
