摘 要:目的探讨宁夏枸杞粗提物对大鼠溃疡性结肠炎的作用。方法采用三硝基苯磺酸(TNBS)-乙醇溶液(含TNBS20~25mg,乙醇浓度30~50%,大鼠TNBS给药剂量80~90mg/kg)建立溃疡性结肠炎大鼠模型,分别给予柳氮磺胺吡啶(SASP)、宁夏枸杞粗提物以及SASP和枸杞粗提物联合用药后,观察各组大鼠一般情况、结肠病变大体形态及病理变化,并检测大鼠血清SOD和结肠组织中MPO活性。结果宁夏枸杞粗提物能明显降低结肠炎大鼠的死亡率,改善一般情况,缩小溃疡面积。枸杞粗提物组、SASP组DAI评分和CMDI评分明显降低,与模型组比较,差异有统计学意义(P〈0.05);模型组大鼠血清SOD和结肠MPO活性与正常组比较,均有统计学差异(P〈0.05);宁夏枸杞粗提物组大鼠血清SOD和结肠MPO活性与模型组相比,均有统计学差异(P〈0.05);与SASP组相比,无统计学差异(P〉0.05);联合用药组大鼠血清SOD和结肠MPO活性与粗提物组及SASP组相比,均有统计学差异(P〈0.05)。结论宁夏枸杞粗提物具有抗大鼠溃疡性结肠炎的作用,可升高大鼠血清中SOD,降低结肠组织中MPO活性;联合用药抗溃疡性结肠炎作用优于单用宁夏枸杞粗提物和SASP,实验结果为进一步研究宁夏枸杞抗溃疡性结肠炎的作用机制提供了实验依据。Objective To investigate the effect of extract of Lycium barbarum L. on ulcerative colitis in rats. Methods UC was induced with combined trinitrobenzene sulfonic acid( TNBS) and ethanol. Rats were randomly divided into five groups: normal group,model control group and SASP group,extract of Lycium barbarum L. group and Combination group. After treatment with medicine,the general conditions of animals and pathological changes in the colon were observed and evaluated. Serum contents levels of SOD and colon tissue levels of MPO were measured by SOD and MPO Detection Kit. Results This study show that the symptoms and pathological changes in the colon were obviously improved in the extract of Lycium barbarum L. group compared to the model control group( P〈0. 05) Serum levels of SOD was significantly decreased( P〈0. 05) in the model control group compared to the normal control group,and colon tissue levels of MPO was significantly increased( P〈0. 05). Serum levels of SOD and colon tissue levels of MPO also differed significantly between the model control group and the extract of Lycium barbarum L.group( P〈0. 05). The extract of lycium barbaruml. group was compared with the SASP group,the difference was not statistically significant( P〉0. 05). Serum levels of SOD in the Combination group was significantly increased( P〈0. 05),and colon tissue levels of MPO was significantly decreased( P〈0. 05) in comparison with those in the extract of Lycium barbarum L. group and SASP group. Conclusion The results suggest extract of Lycium barbarum L. exerts therapeutic effects in UC rats possibly via mechanisms associated with the regulation of serum levels of SOD and colon tissue levels of MPO. The effect of combined medication on rat ulcerative colitis is better than that of single Lycium barbarum L. extract and SASP. The experimental results provide experimental basis for further study on the mechanism of Lycium barbarum anti ulcerative colitis.
