摘 要:目的观察低氧下启膈散和顺铂的抑制食管癌细胞增殖的协同效应,从细胞周期和凋亡探讨其机制。方法制备含药血清,MTT法检测启膈散和顺铂单用或联合应用对食管癌细胞EC9706增殖的影响,流式细胞术检测细胞周期和凋亡。结果 8%、10%浓度的启膈散含药血清与1μg·ml-1顺铂联合抑制EC9706细胞增殖具有明显协同效应,Q值分别为1.18,1.25。用药各浓度与对照组相比细胞G1、G2期比率显著降低,S期显著升高(P<0.01),顺铂高浓度组、启膈散组、联合低浓度组细胞G1期比率高于顺铂低浓度组,S期比率显著低于顺铂低浓度组(P<0.05),联合高浓度组细胞G1期比率明显低于高浓度顺铂组(P<0.05)。除启膈散组外,其他用药各组细胞凋亡率显著高于对照组(P<0.01),联合高浓度组细胞凋亡率高于顺铂高、低浓度组(P<0.05),顺铂组、联合组高浓度组细胞凋亡率明显高于低浓度组(P<0.05)。结论启膈散含药血清和顺铂在抑制食管癌细胞EC9706增殖方面具有协同作用,其机理和诱导细胞凋亡有关。Objective To investigate the synergistic inhibition effect of Qige Powder and cisplatin on proliferation of esophageal carcinoma cells in hypoxia and which mechanism will be explored from Cell cycle and apoptosis. Methods Serum containing drugs was made. The effect of Qige Powder and cisplatin alone or in combination on cells activation was tested by MTT assays. Apoptosis and cell cycle arrest induced by Qige Powder and cisplatin were detected by flow cytometry. Results Qige Powder in the concentration of 8%,10% and 1μg·ml-1cisplatin combination has a synergistic inhibition effect on proliferation of esophageal carcinoma cells,the Q value were 1. 18,1. 25. Compared with the control group,The rate of G1 and G2were significantly lower and the S phase significantly higher in the medication groups. The G1 phase of cisplatin with high concentration group,the Qige Powder Group,and the combination with low concentration group were higher than those in the cisplatin with low concentration group,but the S phase in which was significantly lower than that in the cisplatin low concentration group. The G1 phase of the cells in the combination with high concentration group was significantly lower than that in the high dose cisplatin group. In addition to the group,the apoptosis rate was significantly higher than that in the control group( P〈 0. 01),and the apoptosis rate in the combination with high concentration group was higher than that in the cisplatin with high and low concentration group( P 〈0. 05). The apoptosis rate in the high concentration groups was significantly higher than that in the low concentration groups. Conclusion Qige Powder and cisplatin have synergistic effect on inhibiting proliferation of esophageal carcinoma cell line EC9706,and the mechanism may be related to cell apoptosis induced.
