摘  要:目的探讨中药复方桑瓜饮对实验性2型糖尿病大鼠降血糖作用及其作用机制。方法 SD大鼠高脂饲料喂养,结合腹腔注射链脲佐菌素(STZ)40mg·kg-1,根据血糖值大于11.1mmol·L-1建立糖尿病大鼠模型,成模大鼠随机分为模型组、阳性对照组(二甲双胍0.15g·kg-1)和桑瓜饮组(按生药量计为8.3g·kg-1)。另设正常组。观察给药期间各组血糖(FBG)变化,给药6周后摘眼球取血,检测血清中糖化血红蛋白(Hb A1c)、丙二醛(MDA)、一氧化氮(NO)含量及超氧化物歧化酶(T-SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)活力,并解剖称取新鲜肝脏,测定肝糖原含量。结果桑瓜饮能降低实验性2型糖尿病大鼠FBG和Hb A1c水平,升高肝糖原含量,并提高GSH-Px活力及降低MDA含量,与模型组比较有显著性差异(P<0.05)。结论桑瓜饮对实验性2型糖尿病大鼠模型有降低血糖作用,其降糖机制可能与减少肝糖原分解和提高机体抗氧化能力有关。Objective To investigate the Hypoglycemic Effect and Mechanism of the Sanggua Decoction( SGD) in type 2 diabetic rats. Methods SD rats were constructed by high-fat-diet and combined with intraperitoneal injection of streptozocin 40 mg ·kg-1,whose blood sugar value greater than 11. 1mmol/L is successful DM mold. and the model rats were randomly divided into diabetes model group( T2DM),positive control group( PC)( metformin 0. 15g·kg-1),Sanggua Decoctiongroups( SGD)( raw herb8.3g·kg-1),and the normal control group( NC) was also designed,thechanges of the blood glucose was observed,After 6-week treatment,take the eyeball for blood andseparatethe serum,measure the serumfasting blood glucose( FBG),glycosylated hemoglobin( Hb A1c),MDA,NO,the activtiy of T-SOD,CAT,GSH-Px,then kill the rats and dissect,cut a part of the liver and weigh it,test the amount of hepatic glycol. Results Compared with the model group,SGD couldsignificantly decrease the levels of FBG,Hb A1 c,MDA,increase the amount of hepatic glycol and the activtiy of GSH-Px. Conclusion SGD can effectively reduce the level of FBG. The hypoglycemic mechanism may be related to the reduction of hepatic glycogen decomposition and the growth of the antioxidationin diabetic rats.
