摘 要:目的:研究大鼠灌胃八味沉香散后的药物代谢动力学行为,为该制剂的临床合理用药提供参考。方法:采用HPLC测定大鼠血浆中丁香酚的血药浓度,以3,4-二甲氧基苯乙烯为内标,流动相乙腈(A)-水(B)梯度洗脱(0~5 min,37%~40%A;5~10 min,40%~45%A;10~14 min,45%A;14~23 min,90%A;23~35 min,37%A),检测波长280 nm。将八味沉香散灌胃给药后,利用DAS 2.0软件对大鼠体内丁香酚的平均血药浓度-时间数据进行隔室模型拟合。结果:大鼠经灌胃给药后,以二室模型计算血药浓度与实测值契合较好,优于其他模型。分布半衰期20.031 min,消除半衰期69.315 min。结论:检测血浆中丁香酚含量的方法简便、准确、稳定,适用于八味沉香散中丁香酚的药物代谢动力学分析;丁香酚在大鼠体内分布较快,在大鼠体内主要以消除过程为主,代谢速度中等。Objective: To estimate the pharmacokinetics of eugenol in rats after intragastric administration of Bawei Chenxiang San. Method: HPLC was employed to determine the plasma concentration of eugenol with 3,4-dimethoxystyrene as the internal standard,mobile phase of acetonitrile-water for gradient elution and detection wavelength at 280 nm. After intragastric administration of Bawei Chenxiang San,DAS 2. 0 software conducted compartment model fitting of average plasma concentration-time data of eugenol. Result: After intragastric administration of Bawei Chenxiang San, two-compartment model( weight of 1/cc) calculated the plasma concentration of eugenol was completely consistent with the measured value,and was superior to other models. The distribution half-life( t1/2α) was 20. 031 min and the elimination half-life( t1/2β) was 69. 315 min.Conclusion: The established HPLC method is convenient, accurate and stable, which can be used for pharmacokinetics of eugenol in Bawei Chenxiang San. Eugenol distributes rapidly in rats,the main process of eugenol in rats is elimination with moderate metabolic rate.
