摘  要:研究大鼠迷走神经背核(DMV)和孤束核(NTS)星形胶质细胞在应激性胃黏膜损伤中的作用。将36只雄性Wistar大鼠随机分为对照组和束缚-浸水应激组(RWIS 0.5、1、2、3、5h),通过免疫组织化学技术,检测胶质纤维酸性蛋白(GFAP)的表达。将12只雄性Wistar大鼠随机分为生理盐水对照组和L-AA(星形胶质细胞的抑制剂)组,束缚-浸水应激1 h,检测Fos蛋白和GFAP蛋白的表达及胃黏膜损伤情况。与对照组相比,应激组大鼠在DMV和NTS的不同区段GFAP有不同程度的增加。束缚-浸水应激条件下,星形胶质细胞抑制剂L-AA可显著抑制GFAP的表达及神经元的Fos蛋白的表达,同时胃黏膜损伤明显减轻。星形胶质细胞可能通过调节神经元的激活,参与了束缚-浸水应激,影响了胃肠机能,导致胃黏膜损伤。To investigate the effect of astrocytes in the nucleus of solitary tract( NTS) and dorsal motor nucleus of the vagus( DMV) on the stress-induced gastric mucosal injury in rats. Thirty-six male Wistar rats were randomly divided into the control group and the Restraint water-immersion stress( RWIS)( 0. 5、1、2、3、5h) groups. Glial fibers acidic protein( GFAP) expression were tested by immunohistochemical staining. Twelve male Wistar rats were randomly divided into the saline control group and L-AA( inhibition of astrocytes) group. 1 h after RWIS,the Fos and the GFAP expression and the damage of gastric mucosa were detected.Results showed that compared with the control group,the number of GFAP positive astrocytes in DMV and NTS in the RWIS groups was significantly increased. The GFAP expression peaked at 1 h after the stress and the spatial of GFAP expression were different. L-AA significantly inhibited GFAP and Fos positive expression in the brainstem( NTS and DMV) and reduced gastric mucosal injury.Astrocytes may participate in the gastrointestinal dysfunction and the gastric mucosal damage induced by RWIS through regulating the activation of neurons.
