大鼠肾去交感神经实验动物模型的制备

摘  要:目的 制备大鼠肾去交感神经实验动物模型.方法 52只SPF级SD大鼠随机分为正常对照组(N组)、肾去交感组(RDN组)、心肌梗死组(MI+Sham组)和心肌梗死加肾去交感治疗组(MI+RDN组)四组.MI+Sham组和MI+RSD组分别结扎前降支冠脉制备心梗模型,7d后分别予肾去交感假手术或手术处理.电刺激法评估术前及RDN术后即刻,HE染色法评估术前和MI术后4周肾交感去除情况,ELISA法评估血浆血管紧张素Ⅱ(AngⅡ)、醛固酮(ALD)和去甲肾上腺素(NE)变化情况.结果 RDN术总死亡率为18.1%,手术成功率为81.9%.在RDN术前,肾神经干电刺激可诱导明显交感反应,而术后上述反应消失.HE染色显示,正常未消融的肾动脉外膜外可见完整交感神经,RDN术后3周未见任何完整肾交感神经,肾动脉血管壁未见明显异常.与MI+Sham组相比,MI+RDN组AngⅡ、ALD及NE均明显减少[NE:(669.0±24.0)ng/L比(1349.0±209.0)ng/L,P<0.01;AngⅡ:(205.5±16.4)ng/L比(1069.0±209.1)ng/L,P<0.01;ALD:(290.0±36.0)ng/L比(1049.5±298.5)ng/L,P<0.01].结论 本研究所制备的大鼠肾去交感模型具有良好的操作性、安全性、实用性和有效性.Objective To establish a repeatable and steady rat renal sympathetic denervation (RDN) model. Methods 52 SPF level SD rats were randomly divided into 4 groups, N group, RDN group, MI+Sham group and MI+RSD group. Bilateral renal sympathectomy or sham operation were performed in RDN group and MI+ RSD group or MI+Sham group, separately. Electrical stimulation and HE staining method were used to assess the situation of renal sympathetic denervation at the preoperative and/or postoperative and 4 weeks post-MI, ELISA method was adopted to evaluate the plasma levels of Angiotensin Ⅱ (Ang Ⅱ ), Aldosterone (ALD) and Norepinephrine (NE). Results 22 rats were used for renal sympathetic operation, the total RDN mortality was 18.1% and the RDN success rate was 81.9%. Electrical stimulation induced to obvious sympathetic response pre-operation of RDN, and the above reaction disappeared after RDN. HE staining showed the complete sympathetic nerve outside the outer membrane of the renal arteries, which could not be observed 4 weeks post-RDN, renal artery walls were normal at that time. Compared with MI+Sham group, the plasma levels of Ang Ⅱ , ALD and NE were signif- icantly decreased in MI+RDN group (P〈0.01). Conclusion Rat renal sympathetic animal model made in this study, has a good operability, security, practicability and effectiveness. It can be used to basic studies to evaluate renal sympathetic related diseases.
