摘  要:【目的】建立普萘洛尔微乳凝胶给药后家兔体内血药浓度的侧定方法。【方法】采用高效液相色谱-荧光法测定普茶洛尔血药浓度,用Supelco C18小柱萃取血浆中的普茶洛尔,色谱柱为phenomenex ODS(250 mm×4.6 mm,5μm),流动相为甲醇-0.05 mol/L的碳酸氨水溶液(40:60),流速1.0 ml/min。【结果】普茶洛尔血药浓度在1.0~50.0 ng/ml内线性良好,r2=0.992;不同浓度方法回收率分别为89.93%,93.13%和91.30%。日内和日间精密度RSD分别小于2.56%和5.02%。【结论】建立了简单、快速和准确的家兔体内普茶洛尔经皮血药浓度的固相萃取和高效液相色谱-荧光测定方法。【Objective】To establish a method for determining the microemulsion gel of propranolol in rabbit plasm.【Method】The plasm propranolol was pretreated by a Supelco C18 cartridge and separated on a phenomenex ODS column(250 mm×4.6 mm, 5μm).The mobile phase consisted of methanol 0.05 mol/L ammonium hydrogen carbonate solution(40:60) at a flow-rate of 1.0 ml/min. The plasm concentration of propranolol was determined by HPLC with fluorescence detector.【Result】The plasm concentration of propranolol showed good linearity in the range of 1.0~50.0 ng/ml and the correlation coefficient was 0.992. The recovery rates in different concentrations were respectively 89.93%, 93.13% and 91.30%. The intra-day and inter-day RSDs were less than 2.56% and5.02%.【Conclusion】A simple, rapid and precise method is established for determining the concentration of transdermal propranolol in rabbit plasm by solid phase extraction and HPLC with flurescence detector.
