摘  要:目的 探讨肾去交感神经术(RSD)对心肌梗死大鼠心肌纤维化和转化生长因子-β1(TGF-β1)表达水平的影响.方法 52只雄性SD大鼠随机分为4组:正常对照组(Control组)、肾去交感组(RSD组)、心肌梗死组(MI7d+Sham组)和肾去交感干预组(MI7d+RSD组).MI7d+Sham组和MI7d+RSD组分别给予前降支冠脉结扎术制备心梗模型,7d后行肾交感假手术或去交感术.心脏彩超评估大鼠左心室收缩末期直径(LVESD)、左心室射血分数(LVEF)和左心室短轴缩短率(LVFS).Masson法检测大鼠左心室心肌胶原容积分数(CVF).RT-PCR法和Western-blot法分别检测大鼠左室心肌TGF-β1 mRNA和TGF-β1蛋白的相对表达水平.结果 与MI7d+Sham组相比,M17d+RSD组大鼠LVESD明显减小[LVESD (3.13±0.25)mm比(5.23±0.15)mm,P<0.01],而LVFS和LVEF均增高[LVFS(38.2±4.5)%比(23.1±3.3)%,P<0.01;LVEF(61.7±1.3)%比(40.4±2.7)%,P<0.01].与MI7d+Sham组相比,Mt7d+RSD组胶原容积分数减少[(42.66±7.55)%比(59.33 ±9.79)%,P<0.01],TGF-β1 mRNA(1.47±0.09比2.00±0.09,P<0.01)和TGF-βI蛋白(0.46±0.04比0.73±0.02,P<0.01)的相对表达量均减少.结论 肾去交感神经术能抑制心肌梗死大鼠心肌纤维化并改善心功能,可能与下调TGF-β1的表达有关.Objective To observe the effect of renal sympathetic denervation (RSD) on myocardial fibro- sis and the expression of transforming growth factor-β1 (TGF-β1 ) in myocardial infarction (MI) rats. Methods 52 SPF level SD rats were randomly divided into four groups, control group, RSD group, MI7d+Sham group and MITd+RSD group. Bilateral renal sympathectomy or sham operation were performed in RSD group and MI7d+RSD group or MI7d+Sham group, separately. Ultrasound was adopted to evaluate the changes of left ventricular end- systolic diameter (LVESD), left ventricular fractional shortening (LVFS) and left ventricular ejection fraction (LVEF). Masson staining was used to measure the collagen volume fraction (CVF). RT-PCR and western-blot methods for the detection of myocardial mRNA and protein expression levels of TGF-β1, respectively. Results LVESD was lower in MI7d +RSD group than those in MI7d +Sham group, and LVFS and LVEF were higher [LVESD (3.13±0.25)mm vs (5.23±0.15)mm, P〈0.01, LVFS (38.2±4.5)% vs (23.1±3.3)%, P〈0.01, LVEF (61.7±1.3)% vs (40.4±2.7)%, P〈0.01 ]. Compared with MI7d+Sham group,CVF was lower in MI7d+RSD group [(42.66±7.55)% vs (59.33±9.79)%, P〈0.01], and the relative expression of TGF-β1 mRNA was lower than that in MI7d+Sham group( 1.47±0.09 vs 2.00±0.09, P〈0.01 ), and the relative expression of TGF-β1 protein was lower than that in MI group (0.46±0.04 vs 0.73±0.02, P〈0.01 ). Conclusion Renal sympathetic denervation can inhibit myocardial fibrosis in rats post-MI and improve heart function, which may be achieved by inhibiting the expression of myocardial TGF-β1.
