固相萃取-HPLC法测定人血浆中伏立康唑浓度及其临床应用

摘　　要：目的建立测定人血浆中伏立康唑浓度的固相萃取-HPLC法。方法用Supelclean LC-18 SPE(500 mg)固相萃取小柱提取人血浆中伏立康唑。色谱柱:Shim-Pack C18(4.6 mm×250 mm,5μm);流动相:甲醇-水(57∶43);流速:1.0 m L·min^(-1);进样量:50μL;检测波长:255 nm;柱温:32℃。考察该方法的专属性、标准曲线和定量下限、精密度与回收率、稳定性。结果伏立康唑在0.5~20.0μg·m L^(-1)内线性关系良好,最低定量下限为0.5μg·m L^(-1)(S/N≥3)。伏立康唑的平均提取回收率为88.87%,平均方法回收率为97.17%,平均日内RSD为3.57%,平均日间RSD为4.05%。结论本方法准确、快速、简便,适用于伏立康唑的血浆浓度测定及药代动力学研究。Objective To establish a method for the determination of voriconazole in human plasma by solid phase extraction（SPE） and high performance liquid chromatography（HPLC） and to apply this method for therapeutic drug monitoring of voriconazole.Methods Voriconazole in human plasma was exacted by Supelclean LC-18 SPE（500 mg）.The analytical column was Shim-Pack C18（4.6 mm × 250 mm,5 μm）;the mobile phase consisted of methanol and water（57∶ 43）;the flow rate was1.0 m L·min^（-1）;the injection volumn was 50 μL;the detection wavelength was set 255 nm;and the temperature was 32 ℃.This method was used to detect the plasma concentrations of voriconazole of 10 patients.Results Good liner calibration curve of voriconazole was obtained in the concentration range of 0.5-20.0 μg·m L^（-1）,and the limit of quantitation of voriconazole was 0.5 μg·m L^（-1）（S/N≥3）.Average recovery of extraction was 88.87%.Average method recovery was 97.17%.The RSDs of inter-day and intra-day precision of voriconazole were 3.57%and 4.05%,respectively.Conclusion The established method was accurate,rapid and simple,suitable for clinical therapeutic drug monitoring and pharmacokinetic study.High concentration variability was observed among patients,and routine monitoring of voriconazole may improve the rational administration.
