摘　　要：[目的]通过生物信息学预测分析人TRIM28基因的启动子及蛋白质的理化性质、信号肽、亲疏水性、跨膜区域、蛋白结构、与之相互作用的蛋白质及功能。[方法]使用相应软件分析TRIM28相关信息。[结果]TRIM28的3个启动子中第一个通过甲基化对其表达影响较深;TRIM28蛋白质是由835个氨基酸组成的无信号肽、无跨膜结构的不稳定亲水蛋白质,等电点为5.52,哺乳动物中高度保守;二级结构包括10个α螺旋和12个β折叠片层,三级结构构建需更多可靠的模板;TRIM28主要定位于细胞核,自身及相互作用蛋白质主要参与染色质修饰及DNA损伤修复过程。[结论]TRIM28第一个启动子甲基化影响其表达,蛋白质无信号肽、无跨膜结构、不稳定系数高达46.43,属不稳定亲水蛋白质,定位细胞核,参与染色质修饰和DNA损伤修复。[ Objective ] To analyze the promoter, physicochemical property, signal peptide, transmenbrance domain, hydrophilcity/hydrophobicity, secondary structure, teriary structure, protein - protein interaction and gene ontology of TR1M28 by bioinformation. [ Methods] By meaning of multiple analysis software, the messages of TRIM28 were predicted and analyzed. [ Restilts]The first of 3 promoters was crucial for the expression of TRIM28. And TRIM28 protein was composed of 835 amino acids ,without signal peptide and without transmembrance domain with isoelectric point of 5.52, and it was highly conservative in mammal. There were 10 α - helices and 12 β - sheets in the secondary structure predicted. More reliabletemplates were necessary for 3 D model prediction. TRIM28 located in nucleus. TRIM28 and the interacted proteins took part in chromatin modification and DNA repair. [ Conclusion] The methylation of the first promoter is critical for expression. TRIM28 was a hydrophilic and unstable protein without signal peptide and transmembrance domain. The instability index was up to 46.43. TRIM28 regulated chromatin modification and DNA repair.
