摘 要:目的建立人血浆中多西他赛浓度测定的HPLC方法,用于多西他赛药动学特征的研究。方法色谱柱:Phenomenex C18(250 mm×4.6 mm,5μm);流动相:乙腈-水(60∶40);等度洗脱;流速1 m L·min-1;检测波长230 nm;柱温40℃;进样量20μL;内标:艾司唑仑;血浆样品经3 g·L^(-1)Zn SO4的甲醇溶液处理后,上清液氮气吹干、复溶、进样。结果多西他赛血药浓度检测的线性范围在0.064~4.096 mg·L^(-1)(r>0.999 8),检测限为0.05 mg·L^(-1),方法的回收率为95%~110%,日内、日间RSD均<11%。结论建立的人血浆中多西他赛浓度测定的HPLC方法简便、准确、快速,专属性好,适用于多西他赛血药浓度的常规监测及药动学研究。AIM To establish an HPLC method for the determination of docetaxel in human plasma and investi- gate its clinical pharmaeokinetics. METHODS The analytical column was Phenomenex C18(250 mm×4.6 mm, 5μm) with the mobile phase of aeetonitrile and water (60: 40). The flow rate was 1 mL·min- 1 and the detection wavelength was set at 230 nm. Column temperature was 40℃ and the injection volume was 20 μL. Estazolam was used as internal standard. The plasma samples were pretreated with methanol containing 3 g·L- 1 ZnS04. The supematant was dried with nitrogen, then dissolved and injected. RESULTS The linear range for docetaxel concentration detection was 0. 064 - 4.096 mg· L- 1 ( r 〉 0. 999 8). The detection limit for docetaxel was 0.05 mg·L- 1. The recoveries of the method were between 95%- 110%, and the intra-day precision and inter-day precision (RSD) of analysis were less than 11%. CONCLUSION The method described is simple, sensitive, rapid, and with good specificity. This method can be ap- plied for the plasma eoneentration monitoring of docetaxel and used for pharrnacokinetic study.
