摘  要:目的:研究Aβ25-35寡聚体对原代培养海马神经细胞突触形态及突触蛋白Ng表达的影响。方法:5、10、20umol/L Aβ25一35寡聚体作用于原代培养的海马神经元1h,以免疫细胞化学法检测Ng的表达。结果:与对照组相比:10、20μmol/L Aβ25-35寡聚体致Ng表达减少,差异有显著性(P<0.05,P<0.01),以10μmol/L Aβ25-35组最显著(P<0.01)。结论:Aβ25-35寡聚体对突触有损伤作用与作用浓度有关。Objective: To investigate neurotoxicity in synapses of primary cultured hippocampal neurons induced by Aβ25-35 oligomers. Methods: The hippocampal neurons were incubated by Aβ oligomers with different concentration for 1 hour. Ng was used as index of the measure synaptic damage. The expression of PSD-95 was detected by using immunohistochemical SABC method. Result: Compared with the normal group,the expression of Ng decreased in 1μmol/L Aβ group,5μmol/L Aβ group( P〈0. 05,P〈0. 01). The latter is more significant( P〈0. 01) with statistical significance. Conclusion: Neurotoxicity in Synapse induced by Aβ25-35 oligomers is associated with concentration in a dose-dependent manner.
