水貂出血性肺炎、多杀性巴氏杆菌病、肺炎克雷伯杆菌病三联灭活疫苗的制备及其质量评价研究

摘　　要：为评价水貂出血性肺炎、多杀性巴氏杆菌病、肺炎克雷伯杆菌病三联灭活疫苗的质量,本研究对该三联灭活疫苗进行了疫苗的单剂量、单剂量重复和超剂量免疫的安全性试验,最小免疫剂量、免疫期等效力试验以及疫苗保存期试验。结果表明,该三联灭活疫苗对2月龄以上水貂安全,最小免疫保护剂量为0.5 m L（绿脓杆菌、多杀性巴氏杆菌和肺炎克雷伯杆菌含量分别为3.0×10^9 cfu/m L、3.0×10^9 cfu/m L和2.5×10^9 cfu/m L）,免疫剂量为1.0 m L/只时即可为水貂提供8个月的完全免疫保护,疫苗在2℃-8℃贮藏15个月仍保持原有的免疫保护效率。本研究研制的该三联灭活疫苗使用安全,对受试动物能够提供100%的免疫保护。本研究为水貂出血性肺炎、多杀性巴氏杆菌病和肺炎克雷伯杆菌病的预防和该三联灭活疫苗的正确使用提供了参考依据。In order to develop and evaluate the quality of a tri-combined inactivated vaccine against mink Hemorrhagic pneumonia, Pasteurellosis multocida and Klebsiellosis pneumoniae, the combined vaccine was prepared from the killed Pseudomonas aeruginosa, Pasteurella multocida, Klebsiella pneumoniae, and the vaccine was further evaluated including safety test in minks, minimum immune dose determination by bacteria challenging and the storage duration at different temperatures. The results showed that this vaccine was safe for minks over 2 month-old when inoculated with single dose or double dose or even twice inoculation with single dose of the vaccine. The minimum immune dose of complete protection for minks was 0.5 mL at the concentrations of 3.0×10^9 cfu/mL for P.aeruginosa, 3.0×10^9 cfu/mL for P.multocida and 2.5×10^9 cfu/mL for K.pneumoniae, respectively. The efficient immune protection lasted for 8 months in minks vaccinated with 1.0 mL of the vaccines. The quality of the vaccine kept stable for 15 months in storage at 2-8 ℃. Therefore, the safety and the efficacy of this vaccine was qualified to provide protection to minks against P.aeruginosa, P.multocida or K.pneumoniae infections.
