摘  要:目的制备翻白草Potentilla discolor Bge.提取物银纳米粒,并评价其抗菌活性。方法将翻白草提取物缓慢加入AgNO_3溶液中,离心后沉淀分散于蒸馏水中以制备银纳米粒,并表征其理化性质。以提取时间、提取液用量、AgNO_3溶液用量为影响因素,优化制备工艺。通过测定银纳米粒的抑菌圈直径、抑菌率和最低杀菌浓度(MBC)来考察其抗菌活性。结果最佳条件为提取时间30 min,提取液(生药量)与Ag^+比例1∶0.016 2。翻白草提取物银纳米粒呈近球形,粒径小,分布均匀,性质稳定,对多药耐药型大肠艾希氏菌、金黄色葡萄球菌、加德纳杆菌和白色念珠菌的抑制作用显著高于翻白草提取物、空白银纳米粒和没食子酸银纳米粒(P<0.05,P<0.01),并呈量效关系,而氨苄青霉素和左氧氟沙星无明显抗菌活性。其最低杀菌浓度(MBC)分别为0.5、1.0、1.0、1.5 mg/m L,是空白银纳米粒和没食子酸银纳米粒的2~4倍。结论所建立的翻白草提取物银纳米粒制备工艺稳定合理,在应对多药耐药菌引起的阴道炎有很好的治疗前景。AIM To prepare silver nanoparticles loaded with Potentilla discolor Bge. extract and to evaluate their antimicrobial activities. METHODS The P. discolor extract was slowly added into AgNO3 solution. After centrifugation,the precipitation was dispersed in distilled water for preparing the silver nanoparticles,then the physicochemical properties were characterized. With extraction time,extract amount and AgNO3 solution volume as influencing factors,the preparation was optimized. The antimicrobial activities of silver nanoparticles were determined through the investigation of their antimicrobial circle diameters,inhibition rates and minimal microcidal concentrations. RESULTS The optimal conditions were determined to be 30 min for extraction time,and 1 ∶ 0. 016 2for ratio of extract( crude drug) to Ag-+. The near spherical silver nanoparticles loaded with P. discolor extract,found to be with small particle size,uniform distribution and stable property,exhibited significantly stronger inhibitory effects against multi-resistant Escherichia coli,Staphylococcus aureus,Bacillus gardiner and Candida albicans for vaginitis than those of P. discolor extract,blank silver nanoparticles and gallic acid-loaded silver nanoparticles( P〈0. 05,P〈0. 01) in a dose-dependent manner,while Ampicillin and Levofloxacin displayed no such obvious activities. Their respective minimal microcidal concentrations of 0. 5,1. 0,1. 0 and 1. 5 mg / m L were 2- 4 times of the blank silver nanoparticles and gallic acid-loaded silver nanoparticles. CONCLUSION The P. discolor silver nanoparticles prepared by the aforementioned stable and reasonable preparation process hold more promise to treat vaginitis due to multi-resistant microbes.
