摘 要:【目的】研究新疆塔城地区野生阿魏菇菌株的遗传多样性和遗传分化特征,为阿魏菇驯化选育提供基础材料和科学依据。【方法】利用生物学和ISSR标记技术对23株阿魏菇菌株进行遗传多样性分析。【结果】在阿魏菇菌丝培养特征方面,各菌株除在菌丝颜色上无明显差异外,在菌落形态、菌丝长势等7项特征方面均有明显差异;ISSR标记共扩增出60条DNA条带,多态性条带57条,多态比率为95.00%,供试菌株两两间的相异系数从0.07~0.68,D=0.31时,可将23个供试菌株分为13个类群。【结论】新疆塔城地区野生阿魏菇野生阿魏菇菌株已开始发生遗传分化,并具有较丰富的遗传多样性。【Objective】The genetic diversity of wild P. eryngii strains collected from Tacheng,Xinjiang was estimated,and the purpose was to provide basic materials and scientific basis for the further breeding of ferula mushroom.【Method】Biological characteristics and ISSR markers were used to analyze the genetic diversity of 23 P. eryngii strains.【Result】It was shown that except no obvious difference in color,significant differences of cultural characteristics were observed among tested samples in colonial morphology,mycelium growth vigor and so on. It was shown that the percentage of polymorphic bands from ISSR markers was95. 00%. Cluster analysis showed that the 23 strains of P. eryngii could be divided into 13 groups at the level of D = 0. 31.【Conclusion】The natural populations of P. eryngii from Tacheng has the occurrence of genetic differentiation with relatively rich genetic diversity.
