凉血祛湿止痒汤对小鼠特应性皮炎模型血清IL-2,IL-4及皮损白三烯B_4的影响

摘　　要：目的观察凉血祛湿止痒汤对小鼠特应性皮炎模型血清IL-2,IL-4及皮损白三烯B_4作用效果。方法小鼠特应性皮炎模型造模成功后进行中药凉血祛湿止痒汤(治疗组)干预,并与安慰剂(对照组)进行对照,观察红斑、水肿、表皮剥脱、苔藓化等症状,应用严重度指数评分法评价疗效,应用ELISA法检测血清IL-2,IL-4及皮损白三烯B_4水平。结果以严重度指数评分法比较小鼠特应性皮炎模型红斑、水肿、表皮剥脱以及苔藓化等各项数据,治疗组干预后小鼠特应性皮炎模型皮损明显改善,与本组干预前相比,差异有统计学意义(P<0.01);治疗组干预后与对照组干预后相比差异有统计学意义(P<0.01),治疗组症状评分均优于对照组;对照组有效率为49.18%,治疗组有效率为85.24%,差异有统计学意义(P<0.01),凉血祛湿止痒汤总体疗效优于对照组。治疗组血清IL-4及皮损白三烯B_4浓度明显下降,而血清IL-2显著上升,效果明显优于对照组(P<0.01)。结论中药凉血祛湿止痒汤对小鼠特应性皮炎模型皮损具有控制红斑、消除水肿、促进破损表皮修复、抑制苔藓化的效果,具有提高血清IL-2、拮抗IL-4及皮损白三烯B_4等炎症因子作用。Objective To observe the concentration of Serum IL-2 and IL-4 and Leukotriene B4 of lesions in mice model with Atopic dermatitis（AD）intervented by Liangxuequshizhiyang Soup.Methods Liangxuequshizhiyang Soup（treatment group）intervening atopic dermatitis model after successfully making atopic dermatitis model in mice,which was to be compared with placebo.The change of lesions from erythema, edema, skin exfoliation, and moss were further observed,and the therapeutic effect of Liangxuequshizhiyang Soup was evaluated by the severity index score.The concentration of Serum IL-2 and IL-4 and Leukotriene B4 of lesions were tested in mice model with AD intervented before and after by Liangxuequshizhiyang Soup.Results Analyzed indicators dates from erythema, edema, skin exfoliation, and moss of mice acute eczema model by Severity Index score, Significant improvement in skin lesions of treatment group was compared after intervention with before, the difference was statistically significant（P〈0.01）.At the same time,compared the degree of improvement in skin lesions after intervention of treatment group with control group, the former was better than the latter. The total effective rate of control group was 49.18%, the one of treatment group was 85.24%, the difference was statistically significant（P〈0.01）. The total effect of Liangxuequshizhiyang Soup was superior to the control group.The concentration of Serum IL-4 and Leukotriene B4 of lesions in mice model were decreased significantly, the serum concentration of IL-2 was significant increased in the treatment group.The change of serum IL-4 and Leukotriene B4 of lesions were better than the control group（P〈0.01）.Conclusion Traditional Chinese medicine Liangxuequshizhiyang Soup could control erythema, eliminate edema, repair the damaged epidermis, inhibit moss role of eczema model of skin lesions in mice. At the same time,it can increase the concentration of serum IL-2,and decrease the concentration of serum IL-4 and Leukotriene B4 of lesions.
